hHGF促進(jìn)人骨髓間充質(zhì)干細(xì)胞向肝樣細(xì)胞分化的實(shí)驗(yàn)研究

我國有9300萬的乙型肝炎病毒攜帶者,其中2千萬人為乙肝患者,酒精性肝病及非酒精性肝病的發(fā)病率亦逐年升高,眾多慢性肝臟損傷因素導(dǎo)致纖維結(jié)締組織異常增生,引起肝功能失代償、肝腹水、肝癌、上消化道出血等一系列嚴(yán)重并發(fā)癥,藥物難以醫(yī)治,肝移植是目前治療終末期肝病最為有效的方法,但困于肝源缺乏等問題,極大的限制了其臨床應(yīng)用,骨髓間充質(zhì)干細(xì)胞因其無限增殖及多向分化的潛能,有望成為治療終末期肝病的又一可行方法,并已成為研究熱點(diǎn)。源于中胚層的骨髓間充質(zhì)干細(xì)胞具有極強(qiáng)的可塑造性,多項(xiàng)研究表明,其可分化為三個胚層的細(xì)胞,如成骨細(xì)胞、心肌細(xì)胞、軟骨細(xì)胞、脂肪細(xì)胞、肝細(xì)胞等。在急慢性肝病中,移植骨髓間充質(zhì)干細(xì)胞進(jìn)行臨床治療已經(jīng)在多篇文獻(xiàn)中報道,且有多項(xiàng)實(shí)驗(yàn)已在動物體內(nèi)或體外誘導(dǎo)過程中加入各種生長因子,以達(dá)到提高干細(xì)胞移植療效的目的,目前尚缺乏轉(zhuǎn)染基因后的骨髓間充質(zhì)干細(xì)胞在體外誘導(dǎo)成肝樣細(xì)胞的實(shí)驗(yàn)研究。有研究表明肝細(xì)胞生長因子(HGF)為骨髓間充質(zhì)干細(xì)胞向肝細(xì)胞分化最重要的始動因子之一[8] ,HGF作用的發(fā)揮同細(xì)胞膜受體c-met的結(jié)合密不可分。HGF蛋白與其受體c-Met結(jié)合后導(dǎo)致c-Metβ鏈的酪氨酸T...

第三軍醫(yī)大學(xué)碩士學(xué)位論文density,IOD)值作為結(jié)果 IOD 值,計(jì)算轉(zhuǎn)染前積分光密度與轉(zhuǎn)染后積分光密度的統(tǒng)計(jì)學(xué)差異。結(jié) 果1.骨髓間充質(zhì)干細(xì)胞的培養(yǎng)將細(xì)胞復(fù)蘇后,放置于 37℃,5%CO2 飽和濕度的培養(yǎng)箱中培養(yǎng),首次按 1:2 比例接種在培養(yǎng)瓶,接種后倒置顯微鏡下觀察成圓形的懸浮細(xì)胞,大約于 3h 后細(xì)胞開始貼壁,形態(tài)變?yōu)槿切渭岸噙呅,伸出偽足?4h 后細(xì)胞全部貼壁,由多形變?yōu)殚L梭形、紡錘形,細(xì)胞數(shù)目逐漸增多,細(xì)胞間形成聯(lián)系交織,48h 后細(xì)胞成團(tuán)樣、漩渦、魚群樣生長。連續(xù)培養(yǎng)傳代擴(kuò)增后備用于細(xì)胞轉(zhuǎn)染。
