晚期氧化蛋白產(chǎn)物誘導(dǎo)小腸上皮細胞G1期阻滯的作用及其機制

背景和目的克羅恩病(Crohn’s disease,CD)是一種病因未明的慢性非特異性胃腸道炎癥性疾病。近年來CD發(fā)病率呈逐步增加趨向,因其易反復(fù)發(fā)作,治療花費巨大,給患者、家庭和社會帶來沉重負(fù)擔(dān)。最近的研究發(fā)現(xiàn)小腸上皮細胞G1期阻滯在CD的發(fā)病中具有重要意義。但是,調(diào)控這種細胞周期阻滯的機制仍然不清楚。氧化應(yīng)激是細胞發(fā)生G1期阻滯的原因之一。在氧化應(yīng)激發(fā)生過程中,CD患者體內(nèi)積累了大量的晚期氧化蛋白產(chǎn)物(Advanced oxidation protein products,AOPPs)。因此,本研究旨在闡明AOPPs在調(diào)節(jié)小腸上皮細胞G1期阻滯中所起的作用及其分子機制,為CD的發(fā)生發(fā)展提供新的理論基礎(chǔ),也為CD的防治提供新干預(yù)靶點。方法1.應(yīng)用流式細胞術(shù)分析來自CD病變和正常對照小腸組織的原代小腸上皮細胞周期的分布。2.通過氯胺T法檢測CD患者和志愿者血漿中的絕對AOPPs水平,并通過去除血漿白蛋白含量的差異計算得出相對AOPPs濃度;采用免疫組化染色檢測小腸組織中cyclin E、CDK2和p27kip1的表達情況,并統(tǒng)計分析它們的表達量與CD患者相對AOPPs濃度的相關(guān)性。3....
