細(xì)胞連接蛋白在肝星狀細(xì)胞活化中的作用

[背景]肝硬化是進(jìn)展性肝纖維化的終末期,如果不加以預(yù)防,可能發(fā)展為肝硬化,甚至肝癌和肝衰竭。在肝纖維化的進(jìn)展過程中起著核心作用的是肝星狀細(xì)胞(hepatic stellate cell,HSC)的活化。當(dāng)前的研究主要集中在細(xì)胞因子促活化的作用上,已經(jīng)闡明諸多因子的分子機(jī)制,但是對細(xì)胞因子活化途徑的干預(yù),對于抗纖維化的治療效果無法達(dá)到預(yù)期。而細(xì)胞間接觸和連接是活化后的HSC的重要表現(xiàn),本課題組之前的研究也發(fā)現(xiàn)細(xì)胞接觸與非接觸共培養(yǎng),HSC會表現(xiàn)出不同的活化狀態(tài),因此,肝硬化發(fā)病新的機(jī)制的探索將有助于肝纖維化的治療取得突破。[目的]通過將靜止的肝星狀細(xì)胞與活化的肝星狀細(xì)胞進(jìn)行接觸培養(yǎng)與非接觸共培養(yǎng)進(jìn)行對照,觀察細(xì)胞間連接蛋白CX-43表達(dá)及阻斷后對肝星狀細(xì)胞活化的影響,從而揭示其對HSC活化的作用。[方法]1.通過蛋白酶,膠原酶,后分散液序貫灌注的方法灌注雄性S-D大鼠的肝臟,分散肝臟細(xì)胞并使之消化,再通過密度梯度離心,和細(xì)胞的貼壁時間的差異性,得到分離的肝星狀細(xì)胞,并且量子點標(biāo)記。將分離得到的大鼠的的原代HSC(靜止表型),與HSC-T6細(xì)胞株(活化表型)依照一定的比值接種在提前用Mat...
