ZAG對(duì)棕櫚酸誘導(dǎo)的HepG2細(xì)胞內(nèi)甘油三酯沉積的影響及機(jī)制

目的:構(gòu)建重組人ZAG(Zinc-α2glycoprotein)真核表達(dá)載體,并瞬時(shí)轉(zhuǎn)染HepG2細(xì)胞,觀察HepG2細(xì)胞過表達(dá)ZAG后對(duì)棕櫚酸誘導(dǎo)的甘油三酯沉積的影響。方法:提取HepG2細(xì)胞總RNA,通過RT-PCR方法擴(kuò)增出ZAG序列,克隆入經(jīng)XhoI和SacII雙酶切后的pIRES2-ZsGreen1,重組人ZAG真核表達(dá)質(zhì)粒(pIRES2-ZsGreen1-hZAG)通過核苷酸測序鑒定。使用不同濃度pIRES2-ZsGreen1-hZAG的經(jīng)脂質(zhì)體轉(zhuǎn)染HepG2細(xì)胞24小時(shí),western-blotting鑒定ZAG陽性細(xì)胞,并采用棕櫚酸誘導(dǎo)該細(xì)胞脂肪變性,采用油紅O染色觀察各組細(xì)胞內(nèi)脂滴情況,酶聯(lián)免疫法檢測細(xì)胞內(nèi)甘油三酯的水平。結(jié)果:1.成功構(gòu)建人ZAG基因的真核表達(dá)載體pIRES2-ZsGreen1-hZAG,該載體轉(zhuǎn)染HepG2細(xì)胞后,能顯著增加ZAG蛋白的表達(dá)。2.與對(duì)照組相比,過表達(dá)ZAG的HepG2細(xì)胞能減少棕櫚酸誘導(dǎo)的細(xì)胞內(nèi)甘油三酯含量;油紅O染色結(jié)果顯示過表達(dá)ZAG的HepG2細(xì)胞在棕櫚酸誘導(dǎo)后細(xì)胞內(nèi)橘紅色脂滴較對(duì)照組HepG2細(xì)胞明顯減少。結(jié)論:1.構(gòu)建了Z...
