腹瀉型腸易激綜合征大鼠腸道菌群和腸道屏障功能的變化及相關(guān)分子機制的研究

目的:腹瀉型腸易激綜合征(diarrhea irritable bowel syndrome,IBS-D)是一種臨床常見的功能性腸病,這種疾病的高發(fā)病率嚴(yán)重影響患者的生活質(zhì)量,對衛(wèi)生醫(yī)療資源造成很大的壓力。IBS-D的病理生理學(xué)機制較為復(fù)雜。目前,其發(fā)病機制尚不明確。IBS-D患者常存在明顯的胃腸動力異常,表現(xiàn)出輕度或者嚴(yán)重的腹瀉并伴隨出現(xiàn)腹痛等癥狀。研究發(fā)現(xiàn)腸道屏障功能的破壞以及腸道微生態(tài)的改變是其發(fā)病的重要原因。本實驗旨在探討IBS-D大鼠腸道微生態(tài)的組成改變以及與腸道屏障相關(guān)的緊密連接蛋白的變化,為IBS-D發(fā)病機制提供實驗數(shù)據(jù)和理論基礎(chǔ)。實驗方法:將Wistar雄性大鼠隨機分為對照組和模型組,對大鼠進行胃腸電極埋置手術(shù),分別于胃竇、十二指腸、結(jié)腸部位埋置電極,檢測兩組大鼠胃腸肌電活動的改變。除此之外,對胃腸動力檢測還應(yīng)用了胃排空腸推進和結(jié)腸排珠實驗;模型評價還包括腹壁撤退反射評分(abdominal withdrawal reflex,AWR)實驗、HE染色、大鼠糞便含水量的檢測;采用ELISA技術(shù)檢測血漿中D-乳酸的含量;通過曠場實驗以及糖水偏嗜實驗評價IBS-D大鼠的焦慮抑...
