CCN1/Cyr61在肝硬化—肝癌進程中的作用和機制研究

目的:肝星狀細胞(hepatic stellate cell,HSC)是肝纖維化過程中成纖維細胞的主要來源,肝星狀細胞被激活而轉(zhuǎn)化為肌成纖維樣細胞并分泌細胞外基質(zhì)是肝纖維化發(fā)生、發(fā)展的重要環(huán)節(jié);另外,活化的肝星狀細胞可以促進肝癌細胞生長、遷移和活力,而肝癌細胞反過來也可以促進肝星狀細胞的增殖、遷移、分泌纖維化因子及增強促血管生成的活性,二者相互作用的惡性循環(huán),促進了肝癌的發(fā)展。CCN1即富半胱氨酸61(cysteinerich61, Cyr61),是CCN家族成員之一,是一種重要的細胞外基質(zhì)調(diào)節(jié)因子,通過作用于成纖維細胞,在皮膚創(chuàng)傷-愈合過程中起重要作用。肝臟疾病中,CCN1促進了非酒精脂肪性肝炎中巨噬細胞的浸潤和肝癌細胞的增殖與遷移,在癌周肝硬化和良性肝硬化組織中CCN1呈高表達,而肝星狀細胞膜上高表達CCN1的受體—整合素,提示CCN1可以作用于肝星狀細胞,但作用機制尚不清楚。本研究通過建立肝纖維化動物模型,并結(jié)合細胞共培養(yǎng)和裸鼠皮下瘤等實驗,證明如下假設(shè):CCN1在肝纖維化-肝硬化發(fā)展進程中呈動態(tài)表達,其表達水平與肝纖維化的發(fā)展進程相關(guān);CCN1參與促進肝星狀細胞的活化;肝纖維化...
