TNF-α對兔膽管成纖維細(xì)胞P311/TGF-β1/α-SMA通路的影響及川芎嗪的干預(yù)作用

目的:探討TNF-α對兔膽管成纖維細(xì)胞P311/TGF-β1/α-SMA通路的影響及川芎嗪的干預(yù)作用。方法:分離、培養(yǎng)正常家兔膽管成纖維細(xì)胞并鑒定,將膽管成纖維細(xì)胞分別給予TNF-α、TNF-α聯(lián)合不同濃度的TMP(0.08、0.4、2.0 mg/m L)干預(yù)48 h,以無處理的膽管成纖維細(xì)胞為空白對照,用CCK-8法檢測細(xì)胞增殖水平;real-time PCR檢測細(xì)胞P311、TGF-β1、α-SMA m RNA表達(dá);Western blot檢測細(xì)胞TGF-β1、α-SMA蛋白表達(dá)。結(jié)果:與空白對照比較,膽管成纖維細(xì)胞經(jīng)TNF-α處理后,增殖明顯增強(qiáng),P311、TGF-β1、α-SMA m RNA以及TGF-β1、α-SMA蛋白表達(dá)明顯上調(diào)(均P<0.05);TMP對TNF-α的上述效應(yīng)有抑制作用,并且呈現(xiàn)濃度依耐趨勢,其中0.4、2.0 mg/m L的TMP有明顯作用(均P<0.05)。結(jié)論:TNF-α可能通過調(diào)控P311/TGF-β1/α-SMA信號通路促進(jìn)膽管成纖維細(xì)胞增殖,TMP能抑TNF-α對該通路的活化,故可能對良性膽道狹窄有防治作用。
