Smad3沉默對(duì)IGFBPrP1誘導(dǎo)的肝星狀細(xì)胞作用的研究

研究背景:各種病因可引起肝臟局部炎癥,繼而由此導(dǎo)致的以膠原為主的細(xì)胞外基質(zhì)過度沉積,進(jìn)而形成肝纖維化。肝纖維化是各種慢性肝病發(fā)展至肝硬化的必經(jīng)階段和中心環(huán)節(jié),屬于可逆性病變。近年來,有關(guān)肝纖維化發(fā)病機(jī)制的研究已取得了長足的進(jìn)展,大量研究表明,肝星狀細(xì)胞(hepatic stellate cells, HSC)是細(xì)胞外基質(zhì)(extracellular matrix, ECM)的主要細(xì)胞來源,在肝纖維化形成中起關(guān)鍵性作用。肝纖維化是一個(gè)有多種細(xì)胞因子和多條細(xì)胞信號(hào)通路共同參與的復(fù)雜病理過程。參與肝纖維化的信號(hào)轉(zhuǎn)導(dǎo)通路有:Smad信號(hào)通路、JAK/STAT信號(hào)通路、NF-κB信號(hào)通路以及MAPK信號(hào)通路等,其中Smad信號(hào)通路是目前研究較為清楚的信號(hào)轉(zhuǎn)導(dǎo)通路。有研究顯示,動(dòng)物肝纖維化往往伴隨HSC數(shù)量進(jìn)行性增多,Smad3 mRNA表達(dá)量顯著增高,提示Smad信號(hào)通路在肝纖維化發(fā)生發(fā)展中起著至關(guān)重要的作用。RNA干擾(RNA interference, RNAi)是生物界廣泛存在的保守防御反應(yīng),是近年來發(fā)現(xiàn)的一種重要的轉(zhuǎn)錄后基因沉默方式,它通過一些小的雙鏈RNA阻斷特定基因的表達(dá),誘導(dǎo)mRN...
