搭載klotho蛋白熱敏型聚合凝膠在內(nèi)鏡下治療胃潰瘍

【摘要】:目的:1、探索klotho蛋白是否對GECs(胃上皮細(xì)胞)增殖率和遷移率有促進(jìn)作用;2、制作一款包裹klotho蛋白能用于內(nèi)鏡下的熱敏型聚合凝膠并對材料進(jìn)行評估;3、通過動物實驗驗證聚合凝膠對胃潰瘍的愈合是否有促進(jìn)作用。方法:1、細(xì)胞增殖實驗(EDU)和細(xì)胞遷移實驗(Transell):將對數(shù)期生長的GECs分為klotho蛋白組和對照組,klotho蛋白組添加klotho,觀察兩組細(xì)胞增殖率和遷移率有無差異。2、聚合凝膠的制作及評估:以β-環(huán)糊精包裹klotho蛋白的制成納米微球,用殼聚糖、F-127、甘油等材料制作成一種溫度敏感性凝膠,納米微球與凝膠混合均勻后通過掃描電鏡觀察,并測定生物材料中klotho蛋白的緩釋速度。3、胃潴留實驗:將8只C57小鼠分為2組,對照組和實驗組,分別灌入含1%亞甲藍(lán)的生理鹽水、klotho蛋白凝膠,在0、30、60、120分鐘后取胃,通過活體熒光成像系統(tǒng)檢測熒光強度。4、動物實驗:將60只SD大鼠隨機分為生理鹽水組,空白凝膠組,奧美拉唑組、klotho蛋白凝膠組4組。各組大鼠成模后每天分別用生理鹽水、空白凝膠、奧美拉唑、Klotho蛋白凝膠灌胃1次治療。在治療的第1、3、7天每組隨機處死5只大鼠取材,測量潰瘍面積及新生毛細(xì)血管數(shù)量。潰瘍標(biāo)本切片后行HE染色、PAS染色、PNCA染色,觀察各組組織學(xué)變化情況。結(jié)果:1、細(xì)胞實驗:klotho蛋白組的增值率是對照組的2.3倍,細(xì)胞遷移數(shù)量是對照組的3.6倍。2、電鏡觀察結(jié)果:納米微球直徑在50-200nm之間,可見納米微球勻稱散布在klotho凝膠上面。3、胃潴留實驗:klotho凝膠的熒光的強度在每個時間段都明顯高于對照組。4、動物實驗:klotho凝膠組愈合情況從肉眼觀和組織學(xué)角度都要優(yōu)于各個對照組。結(jié)論:1、證明了Klotho蛋白在酸性條件下能夠促進(jìn)GECs的增殖遷移。2、制備的包裹klotho蛋白凝膠能夠成功用于內(nèi)鏡下并黏附在胃潰瘍部位。3、證實了klotho蛋白凝膠能夠成功地通過灌胃器導(dǎo)管作用于大鼠胃潰瘍部位,修復(fù)潰瘍組織。
