MANF蛋白表達(dá)水平與肝纖維化程度的相關(guān)性研究

【摘要】:目的探討中腦星形膠質(zhì)細(xì)胞源性神經(jīng)營(yíng)養(yǎng)因子(MANF)蛋白在慢性乙型肝炎病毒(HBV)感染患者肝纖維化發(fā)生發(fā)展中的作用及其與臨床特點(diǎn)的相關(guān)性。方法采用相對(duì)和絕對(duì)定量同位素標(biāo)記(ITRAQ)蛋白質(zhì)組學(xué)和免疫組化法檢測(cè)肝臟穿刺組織中MANF蛋白的表達(dá),分析其表達(dá)水平與肝臟炎癥纖維化分期的關(guān)系;采用實(shí)時(shí)熒光定量PCR技術(shù)檢測(cè)正常對(duì)照(NC)、乙肝病毒攜帶者(ASC)、慢性乙型肝炎患者(CHB)及乙肝后肝硬化患者(LC)外周血白細(xì)胞中MANF mRNA的表達(dá)水平,分析其與不同階段慢性HBV感染者的臨床病毒學(xué)和生化學(xué)指標(biāo)的相關(guān)性。結(jié)果 MANF蛋白主要在肝細(xì)胞質(zhì)中表達(dá),其表達(dá)水平隨著肝臟炎癥及纖維化的程度加重而升高。NC組、ASC組和CHB組分別與LC組比較,MANF mRNA的表達(dá)差異均有統(tǒng)計(jì)學(xué)意義(P0.01)。MANF mRNA的表達(dá)水平在乙肝病毒表面抗原(HBsA g)1.5×106IU/L、(1.5×106~2.0×107)IU/L和2.0×107IU/L 3組間差異有統(tǒng)計(jì)學(xué)意義(P0.05);在乙肝病毒e抗原(HBeA g)陽(yáng)性組與陰性組之間差異有統(tǒng)計(jì)學(xué)意義(P0.01);在膽紅素正常組與異常組之間差異有統(tǒng)計(jì)學(xué)意義(P0.01)。結(jié)論 MANF蛋白可能參與了慢性HBV感染者肝纖維化的發(fā)生發(fā)展,并與其臨床特點(diǎn)相關(guān)。
[Abstract]:Objective to investigate the role of mesencephalic astrocyte-derived neurotrophic factor (MANF) protein in the development of hepatic fibrosis in patients with chronic hepatitis B virus (HBV) infection and its correlation with clinical features. Methods relative and absolute quantitative isotope labeling (ITRAQ) proteomics and immunohistochemical method were used to detect the expression of MANF protein in liver puncture tissues, and the relationship between the expression level and the stage of hepatic inflammatory fibrosis was analyzed. The expression of MANF mRNA in peripheral blood leukocytes of patients with (ASC), chronic hepatitis B and posthepatitic cirrhosis of (LC) was detected by real-time fluorescence quantitative PCR. To analyze the correlation between the clinical virology and biochemistry of chronic HBV infection in different stages. Results MANF protein was mainly expressed in the cytoplasm of liver, and its expression level increased with the exacerbation of liver inflammation and fibrosis. NC group, ASC group and CHB group were compared with LC group, respectively. There were statistically significant differences in the expression of MANF mRNA (P0.01). MANF mRNA expression level in (HBsA g) 1.5 脳 106 IUP / L, P 0.01). There was significant difference between (1.5 脳 106) IU/L and 2.0 脳 107IU/L groups (P0.05). There was significant difference between positive and negative groups of hepatitis B virus e antigen (HBeA g) (P0.01), and between normal group of bilirubin and abnormal group (P0.01). Conclusion MANF protein may be involved in the occurrence and development of hepatic fibrosis in patients with chronic HBV infection and may be related to its clinical features.
