去甲腎上腺素對人肝細(xì)胞系表達(dá)瘦素及瘦素受體的影響

【摘要】:目的探討交感神經(jīng)遞質(zhì)去甲腎上腺素(NE)對體外培養(yǎng)人肝細(xì)胞系L02表達(dá)瘦素及瘦素受體的影響。方法用不同濃度的NE作用于體外培養(yǎng)的人肝細(xì)胞系L02,分別于24、48及72 h收集細(xì)胞,Western印跡法檢測其表達(dá)瘦素及瘦素受體蛋白的影響;RT-PCR檢測NE對肝細(xì)胞系L02瘦素及瘦素受體mRNA的影響。結(jié)果①Western印跡結(jié)果顯示,1、10、100μmol/L NE作用于L02細(xì)胞24 h后,瘦素蛋白表達(dá)明顯高于對照組(1.02±0.08,2.24±0.09,2.35±0.12 vs 0.62±0.09,P0.05);瘦素受體蛋白表達(dá)亦明顯高于對照組(1.35±0.13,2.37±0.12,2.39±0.15 vs 0.85±0.13,P0.05)。②RT-PCR檢測L02瘦素以及瘦素受體mRNA的表達(dá)情況,1、10、100μmol/L NE作用于L02 24 h后,瘦素mRNA表達(dá)明顯高于對照組(1.54±0.08,2.37±0.09,2.72±0.12 vs 1.00±0.07,P0.05);瘦素受體mRNA表達(dá)亦明顯高于對照組(1.61±0.08,2.27±0.10,3.39±0.11 vs1.00±0.06,P0.01)。結(jié)論交感神經(jīng)遞質(zhì)NE對體外培養(yǎng)的肝細(xì)胞系瘦素以及瘦素受體的表達(dá)均有促進(jìn)作用,從而參與了肝纖維化進(jìn)程。
[Abstract]:Objective to investigate the effect of norepinephrine (NE), a sympathetic neurotransmitter, on the expression of leptin and leptin receptor in human liver cell line L02 in vitro. Methods Human liver cell line L02 was treated with different concentrations of NE. The expression of leptin and leptin receptor protein was detected by Western blotting. The effects of NE on leptin and leptin receptor mRNA in liver cell line L02 were detected by RT-PCR. Results 1Western blotting showed that the expression of leptin protein in L02 cells was significantly higher than that in control group (1.02 鹵0.08 鹵2.24 鹵0.09 vs, 0.62 鹵0.09 vs, P 0.05). The expression of leptin receptor protein was also significantly higher than that of the control group (1.35 鹵0.13) 2.37 鹵0.12 vs 2.39 鹵0.15 vs 0.85 鹵0.13 (P0.05). The expression of L02 leptin and leptin receptor mRNA was detected by 2RT-PCR, and the expression of L02 leptin and leptin receptor mRNA was detected by 2RT-PCR. The expression of leptin mRNA was significantly higher than that in the control group (1.54 鹵0.08 鹵2.37 鹵0.09 vs, 2.72 鹵0.12 vs, P 0.05). The expression of leptin receptor mRNA was also significantly higher than that of the control group (1.61 鹵0.08 鹵2.27 鹵0.10 鹵3.39 鹵0.11 vs1.00 鹵0.06 vs1.00 P0.01). Conclusion sympathetic neurotransmitter NE can promote the expression of leptin and leptin receptor in liver cell lines in vitro, thus participating in the process of liver fibrosis.
