雙歧桿菌對高脂飲食誘導(dǎo)非酒精性脂肪性肝病大鼠血脂代謝的影響

【摘要】:目的探討雙歧桿菌對高脂飲食誘導(dǎo)非酒精性脂肪性肝病大鼠血脂代謝的影響。方法將45只雄性SD大鼠隨機(jī)等分為對照組、模型組和治療組。對照組喂以普通飼料,模型組喂以高脂飼料,治療組喂以高脂飼料和雙歧桿菌。6 w后比較各組大鼠肝指數(shù)和肝組織學(xué)改變,檢測血清丙氨酸氨基轉(zhuǎn)移酶(ALT)、天冬氨酸氨基轉(zhuǎn)移酶(AST)、總膽固醇(TC)和甘油三酯(TG)的水平以及肝總脂肪(HLs)含量,采用熒光實(shí)時(shí)定量RT-PCR測定禁食誘導(dǎo)脂肪因子(FIAF)mRNA的表達(dá)。結(jié)果模型組肝指數(shù)和肝組織脂肪變性程度均顯著高于對照組(P0.05);而治療組肝指數(shù)和肝組織脂肪變性程度均顯著低于模型組(P0.05)。與對照組比較,模型組ALT、AST、TC、TG和HLs水平顯著升高(P0.05);與模型組比較,治療組ALT、AST、TC、TG和HLs水平顯著降低(P0.05)。與對照組比較,模型組FIAF mRNA的表達(dá)顯著升高(P0.05);與模型組比較,治療組FIAF mRNA的表達(dá)顯著降低(P0.05)。結(jié)論雙歧桿菌能夠降低NAFLD大鼠的血脂水平,減少肝臟組織中脂肪的沉積,改善肝臟的炎癥反應(yīng),其機(jī)制可能與調(diào)節(jié)脂質(zhì)的攝入和抑制FIAF基因的表達(dá)有關(guān)。
[Abstract]:Objective to investigate the effect of Bifidobacterium on lipid metabolism in rats with non-alcoholic fatty liver disease induced by high fat diet. Methods 45 male SD rats were randomly divided into control group, model group and treatment group. The control group was fed with normal diet, the model group with high fat diet, the treatment group with high fat diet and Bifidobacterium. After 6 weeks, the liver index and liver histology of rats in each group were compared. Serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), total cholesterol (TC), triglyceride (TG) and hepatic total fat (HLs) were measured. The expression of adipose factor (FIAF) mRNA induced by fasting was measured by real-time quantitative RT-PCR. Results the liver index and the degree of steatosis in the model group were significantly higher than those in the control group (P0.05), while the liver index and the degree of steatosis in the treatment group were significantly lower than those in the model group (P0.05). Compared with the control group, the levels of ALT,AST,TC,TG and HLs in the model group were significantly higher (P0.05), and the levels of ALT,AST,TC,TG and HLs in the treatment group were significantly lower than those in the model group (P0.05). Compared with the control group, the expression of FIAF mRNA in the model group was significantly increased (P0.05); compared with the model group, the expression of FIAF mRNA was significantly decreased in the treatment group (P0.05). Conclusion Bifidobacterium can decrease the level of serum lipids, decrease the fat deposition in the liver and improve the inflammatory response of the liver in NAFLD rats. The mechanism may be related to the regulation of lipid intake and the inhibition of the expression of FIAF gene.
