重癥急性胰腺炎大鼠回腸肌間神經(jīng)節(jié)血管活性腸肽和一氧化氮合酶陽(yáng)性神經(jīng)元變化

【摘要】:目的:研究伴有胃腸動(dòng)力障礙的重癥急性胰腺炎(SAP)大鼠腸神經(jīng)系統(tǒng)回腸肌間神經(jīng)節(jié)血管活性腸肽(VIP)和一氧化氮合酶(NOS)陽(yáng)性神經(jīng)元的變化,以探討SAP胃腸動(dòng)力障礙的神經(jīng)機(jī)制。方法:SD大鼠20只隨機(jī)分為假手術(shù)組和SAP組,用5%牛黃膽酸鈉逆行胰膽管穿刺注射制作SAP模型。24 h后測(cè)定小腸推進(jìn)比及胰腺病理評(píng)分。制作回腸肌間神經(jīng)節(jié)全層標(biāo)本,應(yīng)用雙重免疫熒光染色,用熒光顯微鏡觀察VIP和NOS陽(yáng)性神經(jīng)元的形態(tài)及計(jì)算其占總神經(jīng)元的百分比。結(jié)果:與假手術(shù)組相比,SAP組大鼠腹腔腹水形成,腸系膜見鈣化灶,腸管明顯擴(kuò)張,小腸推進(jìn)比顯著降低[(51.37±5.04)%vs.(30.20±3.82)%,t=10.570,P0.01],胰腺病理評(píng)分增高(3.74±0.23 vs.13.78±0.65,t=-45.858,P0.01),VIP和NOS陽(yáng)性神經(jīng)元的比例明顯增高[(8.48±1.06)%vs.(14.22±2.12)%,t=7.642,P0.01;(29.04±3.36)%vs.(40.55±2.59)%,t=-8.570,P0.01]。結(jié)論 :SAP胃腸動(dòng)力障礙可能與VIP和NOS兩種抑制性神經(jīng)元的表達(dá)上調(diào)相關(guān)。
[Abstract]:Aim: to study the changes of vasoactive intestinal peptide (VIP) and nitric oxide synthase (NOS) (NOS) positive neurons in ileal intermuscular ganglion (IGM) of severe acute pancreatitis (SAP) rats with gastrointestinal motility disorder (SAP) in order to explore the neural mechanism of SAP gastrointestinal motility disorder. Methods Twenty male Sprague-Dawley rats were randomly divided into sham operation group and SAP group. The SAP model was made by retrograde injection of 5% sodium taurocholate into the pancreatopancreatopancreatic duct. The small bowel propulsion ratio and pancreatic pathological score were measured after 24 hours.
