【摘要】：目的探討感染后腸易激綜合征(PI-IBS)小鼠不同腸段腸黏膜熱休克蛋白70(HSP70)的表達(dá),及其與炎癥細(xì)胞因子表達(dá)的相關(guān)性。方法旋毛蟲(chóng)感染小鼠,不同腸段腸黏膜組織蘇木精-伊紅染色法(HE)染色,炎癥評(píng)分。腹壁撤退反射(AWR)和結(jié)腸傳輸試驗(yàn)(CTT)觀察內(nèi)臟敏感性和腸道動(dòng)力變化。免疫組織化學(xué)及酶聯(lián)免疫吸附試驗(yàn)(ELISA)檢測(cè)不同腸段HSP70和炎癥細(xì)胞因子的表達(dá)。Spearman相關(guān)性分析HSP70與炎癥細(xì)胞因子表達(dá)的相關(guān)性。結(jié)果 1小鼠感染旋毛蟲(chóng)后末段回腸和結(jié)腸出現(xiàn)明顯炎癥,炎癥消退后仍存在明顯的內(nèi)臟高敏感和腸道動(dòng)力異常。2PI-IBS小鼠空腸和末段回腸的HSP70表達(dá)較對(duì)照組明顯增強(qiáng)(P0.01)。3PI-IBS小鼠末段回腸白細(xì)胞介素-17(IL-17)、干擾素-γ(IFN-γ)水平明顯升高,空腸、末段回腸及結(jié)腸白細(xì)胞介素-10(IL-10)水平明顯降低(P0.01)。4PI-IBS小鼠空腸HSP70表達(dá)與炎癥細(xì)胞因子IL-10呈負(fù)相關(guān)(P0.01);末段回腸HSP70表達(dá)與IL-10呈負(fù)相關(guān)(P0.01),與IL-17、IFN-γ呈正相關(guān)(P0.01)。結(jié)論 1不同腸段HSP70和炎癥細(xì)胞因子的相關(guān)差異性表達(dá)可能在PI-IBS發(fā)病中起作用。2末段回腸可能是PI-IBS最重要的炎癥免疫啟動(dòng)和調(diào)控部位。
[Abstract]:Objective to investigate the expression of heat shock protein 70 (HSP70) in different intestinal segments of irritable bowel syndrome (PI-IBS) mice and its correlation with inflammatory cytokines. Methods the intestinal mucosa of mice infected with Trichinella spiralis was stained by hematoxylin-eosin staining and inflammatory score. Abdominal withdrawal reflex (AWR) and colonic transit test (CTT) were used to observe the visceral sensitivity and intestinal motility. Immunohistochemistry and enzyme linked immunosorbent assay (ELISA) were used to detect the expression of HSP70 and inflammatory cytokines in different intestinal segments. The correlation between HSP70 and inflammatory cytokines was analyzed. Results 1the ileum and colon of mice infected with Trichinella spiralis showed obvious inflammation. The expression of HSP70 in jejunum and ileum of mice with abnormal intestinal motility and visceral hypersensitivity after inflammation subsided was significantly higher than that in control group (P0.01) .3The level of Interferon- 緯 (IFN- 緯) and jejunum were significantly increased compared with the control group (P0.01). The levels of interleukin-10 (IL-10) in the ileum and colon decreased significantly (P0.01) .4The expression of HSP70 in jejunum was negatively correlated with the inflammatory cytokine IL-10 (P0.01), and the expression of HSP70 in the terminal ileum was negatively correlated with IL-10 (P0.01) and positively correlated with IL-17,IFN- 緯 (P0.01). Conclusion 1 the differential expression of HSP70 and inflammatory cytokines in different intestinal segments may play an important role in the pathogenesis of PI-IBS. 2. The ileum may be the most important inflammatory immune priming and regulating site of PI-IBS.
