益生菌對(duì)潰瘍性結(jié)腸炎大鼠腸黏膜TLRs表達(dá)的影響

【摘要】:目的:通過(guò)建立潰瘍性結(jié)腸炎大鼠模型,探討益生菌對(duì)潰瘍性結(jié)腸炎(UC)大鼠模型腸黏膜Toll樣受體(TLRs)表達(dá)的調(diào)控,及其在潰瘍性結(jié)腸炎(UC)發(fā)生發(fā)展中可能的作用機(jī)制。方法:本實(shí)驗(yàn)分為正常對(duì)照組、模型組、金雙歧治療組、Toll樣受體4單克隆抗體(TLR4mAb)干預(yù)組。采用三硝基苯磺酸(TNBs)法造模。對(duì)每組大鼠進(jìn)行疾病活動(dòng)指數(shù)(DAI)評(píng)分及組織病理學(xué)(HPS)評(píng)分,采用實(shí)時(shí)熒光定量(RT-PCR)法測(cè)定TLR4、TLR2mRNA的表達(dá),酶聯(lián)免疫吸附試驗(yàn)(ELISA)檢測(cè)血清TNF-α、IL-1β的濃度。結(jié)果:模型組結(jié)腸組織TLR4、TLR2mRNA的表達(dá)及血清TNF-α、IL-1β的濃度均明顯高于正常對(duì)照組,金雙歧及TLR4mAb干預(yù)后,其表達(dá)均明顯低于模型組,差異有統(tǒng)計(jì)學(xué)意義。結(jié)論:潰瘍性結(jié)腸炎的發(fā)生與異常免疫反應(yīng)有關(guān)。益生菌可改善潰瘍性結(jié)腸炎大鼠的結(jié)腸炎癥反應(yīng),其機(jī)制可能與影響腸黏膜TLRs表達(dá)及炎癥因子的水平有關(guān)。
[Abstract]:Objective: to investigate the regulation of probiotics on the expression of Toll like receptor (TLRs) in intestinal mucosa of rats with ulcerative colitis (UC) by establishing a rat model of ulcerative colitis and its possible mechanism in the pathogenesis and development of ulcerative colitis. Methods: the experiment was divided into normal control group, model group and TLR4mAb intervention group. The model was made by (TNBs) method with trinitrobenzene sulfonic acid. The disease activity index (DAI) score and histopathological (HPS) score were measured in each group. The expression of TLR4,TLR2mRNA was measured by real-time fluorescence quantitative (RT-PCR) method, and the concentration of serum TNF- 偽 -1 尾 was detected by Elisa (ELISA). Results: the expression of TLR4,TLR2mRNA in colonic tissue and the concentration of serum TNF- 偽 -1 尾 in the model group were significantly higher than those in the normal control group. After the intervention of Jin Bifidobacterium and TLR4mAb, the expression levels were significantly lower in the model group than in the model group, and the difference was statistically significant. Conclusion: the occurrence of ulcerative colitis is related to abnormal immune response. Probiotics can improve the colitis response in rats with ulcerative colitis. The mechanism may be related to the influence of intestinal mucosal TLRs expression and inflammatory factor levels.
