辣椒素預(yù)防吲哚美辛致急性胃黏膜損傷的作用及機(jī)制

【摘要】:目的:研究不同時(shí)期辣椒素(CAP)對吲哚美辛致急性胃黏膜損傷的作用及機(jī)制。方法:成年雄性健康SD大鼠80只,隨機(jī)分為8組,每組10只,實(shí)驗(yàn)分兩期,Ⅰ期為2周,Ⅱ期為4周。Ⅰ期分別為A1組(空白對照組)、B1組(吲哚美辛模型組)、C1組(CAP預(yù)先給藥組)、D1組(CAP預(yù)先給藥+吲哚美辛模型組);Ⅱ期分組方法同I期。采用吲哚美辛復(fù)制大鼠胃黏膜損傷模型,于應(yīng)激4 h后處死大鼠,收集胃液測定胃酸總酸度,肉眼計(jì)數(shù)胃黏膜損傷指數(shù),行胃竇組織病理學(xué)觀察,檢測胃黏膜辣椒素受體(TRPV1)、降鈣素基因相關(guān)肽(CGRP)的表達(dá),檢測胃竇組織丙二醛(MDA)、超氧化物歧化酶(SOD)及前列環(huán)素(PGI2)的水平。結(jié)果:I期:A1組、C1組胃黏膜未見損傷;與B1組相比,D1組胃黏膜損傷程度、胃酸總酸度、MDA值差異無統(tǒng)計(jì)學(xué)意義(P0.05),SOD、PGI2值顯著升高(P0.05),TRPV1、CGRP表達(dá)顯著增多(P0.05)。Ⅱ期:A2組、C2組胃黏膜未見損傷;與B2組相比,D2組胃黏膜損傷程度顯著減輕(P0.05),胃酸總酸度顯著降低(P0.05),MDA值顯著降低(P0.05),SOD、PGI2值顯著升高(P0.05),TRPV1、CGRP表達(dá)顯著增多(P0.05)。結(jié)論:(1)CAP 1 mg/(kg·d)連續(xù)灌胃2、4周,對正常大鼠胃黏膜大體形態(tài)、病理組織學(xué)無損傷作用;(2)預(yù)先給予CAP 1 mg/(kg·d)連續(xù)灌胃4周具有預(yù)防吲哚美辛致大鼠急性胃黏膜損傷的作用。
