中藥高位保留灌腸對STC大鼠結(jié)腸ICC與血清VIP的影響研究

【摘要】:目的:觀察通腑潤腸湯高位保留灌腸治療慢傳輸型便秘(STC)模型大鼠的效果及對結(jié)腸Cajal間質(zhì)細胞(ICC)、血管活性腸肽(VIP)的影響,探討通腑潤腸湯高位保留灌腸治療STC的效果及作用機制。方法:將60只SD大鼠隨機分為A組(空白對照組)、B組(便秘模型組)、C組(莫沙必利組)、D組(中藥高位保留灌腸低劑量組)、E組(中藥高位保留灌腸中劑量組)、F組(中藥高位保留灌腸高劑量組)。造模期間用復(fù)方地芬諾酯對B組、C組、D組、E組、F組灌胃造模,用生理鹽水對A組灌胃。造模成功后,A組、B組用生理鹽水灌胃,剩余各組用對應(yīng)藥物治療。觀察各組大鼠造模前、造模成功后及治療完成后的一般情況、24h內(nèi)的糞便粒數(shù)及糞便的含水率,治療結(jié)束后檢測SD大鼠的結(jié)腸炭末推進率、血清VIP濃度、結(jié)腸ICC數(shù)量。結(jié)果:(1)在排便粒數(shù)上,C、D、E、F組和B組比較的p值均為0.000,P0.05;E組和C組、D組比較的p值均為0.001,p0.05;F組和C組、D組比較的p值均為0.001,p0.05;C組、D組比較比較的p值0.930,p0.05;E組、F組比較的p值為0.784,p0.05。(2)在糞便含水率上,B組和C組比較的p值為0.820,p0.05;B和D、E、F組比較的p值為依次為0.002、0.001、0.001,p0.05;D組和E、F組比較的p值依次為0.721、0.752,p0.05;E組和F組比較的p值為0.960,p0.05;(3)在結(jié)腸炭末推進率上,B組和C、D、E、F組比較的p值都為0.000,p0.05;E組和C、D組比較的p值都為0.000,p0.05;F組和C、D組比較的p值都為0.000,p0.05;C組和D組比較的p值為0.710,p0.05;E組和F組比較的p值為0.964,p0.05。(4)在影響VIP濃度上,C、D、E、F組和B組比較的p值都依次為0.000、0.001、0.000、0.000,P0.05;而C組和D、E、F組比較的p值依次為0.593、0.954、0.643;D組和E、F組比較的p值依次為0.642、0.933;E組和F組比較的p值為0.694,p0.05。(5)在影響ICC數(shù)量上,C、D組與B組比較的p值依次為0.311、0.106,P0.05;E、F組與B組比較的p值依次0.006、0.020,p0.05。結(jié)論:通腑潤腸湯高位保留灌腸通過增加結(jié)腸ICC數(shù)量、血清VIP濃度來增加排便量、提高糞便含水率、促進腸道傳輸以改善便秘癥狀。通腑潤腸湯高位保留灌腸中、高劑量療效相當(dāng),均較通腑潤腸湯高位保留灌腸低劑量及莫沙必利治療效果好。
[Abstract]:Objective: to observe the effect of Tongfu Runchang decoction high position retention enema on slow transit constipation (STC) model rats and the effect of Tongfu Runchang decoction on (ICC), vasoactive intestinal peptide (VIP) in interstitial cells of colon Cajal. To explore the effect and mechanism of Tongfu Runchang decoction (Tongfu Runchang decoction) in the treatment of STC with high retention enema. Methods: sixty Sprague-Dawley rats were randomly divided into two groups: group A (control group), group B (constipation model group), group C (mosapride group), group D (low dose group of high retention enema of traditional Chinese medicine) and group E (middle dose group of high retention enema of traditional Chinese medicine). During the course of modeling, compound diphenoxylate was used to make models in group B, group C, group D, group C, group E, group F, and group A, with normal saline. The rats in group A were given normal saline and the rest were treated with corresponding drugs. The number of fecal particles and the fecal moisture content were observed before, after and after treatment. The colon carbon propelling rate, serum VIP concentration and colonic ICC content of SD rats were measured after treatment. Results: (1) the p values of group F and group B were both 0.000, P0.05E and C, respectively. The p values of group F and group C were both 0.001p0.05F and 0.001p0.05C, respectively. The p values of group D and group D were 0.930 and 0.05g respectively. (2) the p values of group B and group C were 0.820, p0.05B and group F, respectively. The p values of group D and group E were 0.7210.752p0.05E and 0.960p0.05E, respectively; (3) the rate of propelling of colon carbon in group B and group C were 0.960p0.05; (3) in group B and group C, the p values of group B and group C were 0.960p0.05; (3) the p values of group B and group C were respectively 0.7210.752p0.05E and 0.960p0.05e; (3) the p values of group B and group C were higher than those of group F and group B (P = 0.960p0.05) respectively; (3) the p values of group B and group C were 0.960p0.05 respectively. (4) the difference of VIP concentration between group F and group B was found to be higher than that of group C and group D (P = 0.605), respectively. Conclusion: Tongfu Runchang decoction high retention enema can increase fecal excretion, increase fecal moisture and improve intestinal transport to improve constipation symptoms by increasing the number of colon ICC and serum VIP concentration. In the high retention enema of Tongfu Runchang decoction, the effect of high dose retention enema was equal, which was better than that of Tongfu Runchang decoction high retention enema low dose and mosapride.
