遠隔缺血后適應(yīng)對大鼠小腸缺血再灌注損傷的保護作用

【摘要】:目的探討遠隔缺血后適應(yīng)是否對大鼠腸道缺血再灌注損傷起保護作用。方法健康雄性昆明種大鼠24只,隨機分為假手術(shù)組(Con組)、缺血再灌注組(ischemia reperfusion,IR組)和遠隔缺血后適應(yīng)組(remote ischemic postconditioning,RIP組)共3組,每組8只。Con組只給予解剖游離腸系膜上動脈;IR組解剖出腸系膜上動脈,在腸系膜上動脈缺血60min后再灌注120min;RIP組在腸系膜上動脈缺血20min后捆綁左下肢形成4個間隔5min的循環(huán)缺血再灌注(共40min),再進行再灌注2h。測定回腸組織中丙二醛(maleiedialdehyde,MDA)和髓過氧化物酶(myeloperoxidase,MPO)的含量;光鏡下觀察腸道組織形態(tài)學改變,對小腸黏膜損傷程度進行評分。結(jié)果光鏡下觀察IR和RIP組有明顯的腸道損傷,RIP組腸道損傷程度明顯較IR組輕。小腸損傷按Chiu氏評分,RIP組Chiu評分值(2.10±0.63)明顯低于IR組(3.20±0.76)(P0.05)。IR和RIP組回腸組織中MDA和MPO含量明顯高于Con組(均P0.05),而RIP組回腸組織中MDA及MPO含量明顯低于IR組(均P0.05)。結(jié)論遠隔缺血后適應(yīng)對小腸缺血再灌注損傷具有顯著保護作用,這種遠隔器官缺血后適應(yīng)可能是通過減輕活性氧損傷及炎癥反應(yīng)來保護腸道。
[Abstract]:Objective to investigate the protective effect of adaptation after distant ischemia on intestinal ischemia reperfusion injury in rats. Methods Twenty-four healthy male Kunming rats were randomly divided into three groups: sham operation group (Con group), ischemia reperfusion group (IR group) and (remote ischemic postconditioning group (remote ischemic group). The superior mesenteric artery was dissected by dissecting the superior mesenteric artery in the IR group, and the superior mesenteric artery was dissected in the IR group. After superior mesenteric artery ischemia (60min) and reperfusion 120 minutes after reperfusion, the left lower extremities were bound to form four 5min circulatory ischemia reperfusion (40min) after superior mesenteric artery ischemia (20min), and then reperfusion was performed for 2 h. The contents of malondialdehyde (MDA) and myeloperoxidase (MPO) in ileum tissue were measured and the intestinal tissue morphology was observed under light microscope to evaluate the degree of intestinal mucosal injury. Results the degree of intestinal injury in RIP and IR group was significantly less than that in IR group. The levels of MDA and MPO in ileum of RIP group were significantly higher than those of Con group (P0.05), and the contents of MDA and MPO in RIP group were significantly lower than those in IR group (P0.05). Conclusion adaptation after remote ischemia has a significant protective effect on intestinal ischemia-reperfusion injury. This adaptation may protect the intestinal tract by reducing reactive oxygen damage and inflammatory reaction.
