【摘要】：目的通過(guò)構(gòu)建小鼠感染模型,研究幽門(mén)螺桿菌(Hp)感染對(duì)白細(xì)胞相關(guān)免疫球蛋白樣受體-1(LAIR-1)表達(dá)的影響。方法將32只C57BL/6小鼠隨機(jī)分為對(duì)照組和感染組。感染組通過(guò)Hp菌液灌胃建立小鼠感染模型,對(duì)照組以PBS灌胃。6周后處死小鼠,采用胃黏膜分離培養(yǎng)、革蘭染色、PCR及生化反應(yīng)檢查Hp定植情況,采用流式細(xì)胞術(shù)檢測(cè)小鼠外周血單個(gè)核細(xì)胞(PBMC)中單核細(xì)胞和淋巴細(xì)胞表面LAIR-1的表達(dá)。結(jié)果感染組小鼠Hp感染率為91.3%(21/23)。感染組和對(duì)照組小鼠PBMC中單核細(xì)胞比例分別為(22.85±3.56)%和(49.85±0.35)%,淋巴細(xì)胞比例分別為(40.93±10.05)%和(30.15±0.35)%,差異均有統(tǒng)計(jì)學(xué)意義(t=5.845和2.137,P0.05)。感染組和對(duì)照組小鼠PBMC表面LAIR-1表達(dá)率分別為(70.16±7.75)%和(83.4±0.1)%,淋巴細(xì)胞表面LAIR-1表達(dá)率分別為(56.81±11.37)%和(72.3±0.6)%,差異均有統(tǒng)計(jì)學(xué)意義(t=3.761和2.491,P0.05)。結(jié)論 Hp感染下調(diào)了小鼠PBMC中單核細(xì)胞及LAIR-1的表達(dá),上調(diào)淋巴細(xì)胞比例但下調(diào)其LAIR-1的表達(dá)。
[Abstract]:Objective to study the effect of Helicobacter pylori (Hp) infection on the expression of leukocyte related immunoglobulin like receptor -1 (LAIR-1) by constructing a mouse infection model. Methods 32 C57BL/6 mice were randomly divided into the control group and the infection group. The infection group was injected with Hp bacteria to establish the mice infection model, and the control group died of the mice after.6 weeks after PBS gavage. The expression of LAIR-1 in mononuclear cells and lymphocytes in peripheral blood mononuclear cells (PBMC) of mice was detected by flow cytometry. The infection rate of Hp in infected mice was 91.3% (21/23). The proportion of mononuclear cells in PBMC of the infected group and the control group was respectively (PBMC), and the ratio of mononuclear cells in PBMC of the infected group and the control group was respectively (Hp). 22.85 + 3.56% and (49.85 + 0.35)%, and the proportion of lymphocytes were (40.93 + 10.05)% and (30.15 + 0.35)% respectively. The difference was statistically significant (t=5.845 and 2.137, P0.05). The LAIR-1 expression rate of PBMC on the surface of the infected and control mice was (70.16 + 7.75)% and (83.4 + 0.1)% respectively, and the LAIR-1 expression rate of the lymphocyte surface was (49.85)% and The difference was statistically significant (t=3.761 and 2.491, P0.05). Conclusion Hp infection downregulated the expression of mononuclear cells and LAIR-1 in mouse PBMC, up - regulation of lymphocyte proportion but down the expression of LAIR-1.
