文章摘要:目的:探討NADPH氧化酶Noxl和Duox2在小鼠腸炎發(fā)病中的表達(dá)及意義.方法:將6-8周齡的C57BL/6,小鼠隨機(jī)分為正常組、1.5%葡聚糖硫酸鈉(dextran sulphate sodium,DSS)組和3.0%DSS組,每組10只,正常組正常飲水,1.5%和3.0%DSS組分別自由飲用含有1.5%或者3.0%DSS的飲水,共6d建立小鼠急性腸炎模型.通過疾病活動(dòng)指數(shù)(disease activity index,DAI)評(píng)分、結(jié)腸長度測定和HE染色等方法評(píng)估腸道炎癥程度.利用生化方法檢測血清丙二醛(malondialdehyde,MDA)含量以及結(jié)腸組織中氧化型谷胱甘肽/還原型谷胱甘肽(oxidized glutathione/glutathione,GSSG/GSH)比率等氧化應(yīng)激指標(biāo),以評(píng)估機(jī)體氧化應(yīng)激程度.采用免疫組織化學(xué)方法和實(shí)時(shí)定量PCR技術(shù)分別檢測小鼠結(jié)腸組織中NADPH氧化酶Nox1和Duox2蛋白及mRNA表達(dá)情況.結(jié)果:正常組小鼠無腸炎,1.5%DSS組、3.0%DSS組小鼠分別呈輕度和重度腸炎.氧化應(yīng)激指標(biāo)(MDA、GSSG/GSH)在1.5%DSS組升高,而3.0%DSS組進(jìn)一步升高(均P〈0.05).NADPH氧化酶Nox1與Duox2蛋白和mRNA在不同炎癥程度時(shí)表達(dá)不同:Nox1蛋白和mRNA在正常組呈高表達(dá),在1.5%DSS組表達(dá)下調(diào)(P〈0.05),在3.0%DSS組進(jìn)一步下調(diào)(P〈0.05);Duox2蛋白和mRNA在1.5%DSS組表達(dá)較正常組明顯上調(diào)(P〈0.05),而在3.0%DSS組表達(dá)下調(diào)至正常水平.結(jié)論:Nox1主要在維持腸道正常生理功能中發(fā)揮作用,而Duox2除了維持正常生理功能外,可能還積極地參與腸炎發(fā)病過程.
Abstr:AIM:To investigate the significance of expression of NADPH oxidases Nox1 and Duox2 in the pathogenesis of mouse colitis.METHODS:Six-to-eight-week-old C57BL/6mice were randomly divided into a control group,a 1.5%dextran sulfate sodium(DSS)group and a 3%DSS group(n- 10 for each group).Colitis was induced in mice by giving1.5%DSS or 3%DSS in drinking water for 6d,and mice in the control group was given drinking water only.Disease activity index(DAI),colon length and inflammatory score of the colon were observed.Oxidative stress indicators,malondialdehyde(MDA) in serum and oxidized glutathione/glutathione(GSSG/GSH) ratio in colon tissue,were measured by biochemical methods.The protein and mRNA expression of Noxl and Duox2 in colon tissue of mice was evaluated by immunohistochemistry and quantitative real-time PCR,respectively.RESULTS:There was no colitis in the control group,while mild and severe enteritis was found in mice in the 1.5%DSS group and3.0%DSS group,respectively.Oxidative stress(MDA and GSSG/GSH) was enhanced along with the increased concentration of DSS(P 0.05 for both).The expression of Noxl and Duox2 protein and mRNA was different with the severity of inflammation:Noxl protein and mRNA was highly expressed in normal colon epithelial cells,but down-regulated in the 1.5%DSS group(P〈 0.05),and further reduced in the 3%DSS group(P 〈0.05);Duox2 protein and mRNA expression was increased(P〈 0.05)in mice in the 1.5%DSS group compared to the control group,but returned to the control level in the 3%DSS group.CONCLUSION:Noxl is mainly involved in the functional and structural maintenance of normal colonic epithelium,while Duox2 may actively participate in the inflammatory pathogenesis besides the physiological process of the colon.
