【摘要】：目的在細(xì)胞水平上分析乙型肝炎病毒(HBV)復(fù)制及其編碼的X基因(HBx)表達(dá)對(duì)細(xì)胞基因表達(dá)譜的影響,特別是免疫相關(guān)基因表達(dá)水平的變化。方法通過慢病毒和pcDNA瞬時(shí)轉(zhuǎn)染過表達(dá)HBx基因,利用RNA-Seq和RT-qPCR等方法檢測(cè)免疫相關(guān)基因的表達(dá)水平,并在HBV復(fù)制細(xì)胞系中進(jìn)行驗(yàn)證。結(jié)果 HBV復(fù)制和HBx表達(dá)可以劑量依賴性抑制免疫檢查點(diǎn)程序性死亡分子1(PD-1)配體基因(PD-L1/CD274)的表達(dá),而HBx基因的H-box突變體的表達(dá)失去該抑制效應(yīng)。結(jié)論HBV/HBx具有抑制PD-L1/CD274基因表達(dá)的能力,在病毒感染的急性期解除抗原特異性T細(xì)胞激活的檢查點(diǎn),活化細(xì)胞毒性T細(xì)胞,這可能導(dǎo)致T細(xì)胞對(duì)高度復(fù)制的細(xì)胞進(jìn)行攻擊和清除,幫助病毒進(jìn)入較低復(fù)制狀態(tài),達(dá)到病毒-宿主的平衡狀態(tài)并奠定HBV慢性感染的基礎(chǔ)。
[Abstract]:Objective to investigate the effect of HBV replication and its X gene expression on cell gene expression profile at cellular level, especially the changes of immune-related gene expression level. Methods the expression level of immune-related genes was detected by RNA-Seq and RT-qPCR, and verified in HBV replication cell line by transient transfection of HBx gene by lentivirus and pcDNA. Results HBV replication and HBx expression could inhibit the expression of PD-L1 / CD274) in a dose-dependent manner, while the expression of H-box mutants of HBx gene lost the inhibitory effect. Conclusion HBV/HBx has the ability to inhibit the expression of PD-L1/CD274 gene, deactivate the checkpoint of antigen-specific T cell activation and activate cytotoxic T cells in the acute phase of viral infection, which may lead to T cells attacking and clearing highly duplicated cells. To help the virus into a low replication state, to achieve a virus-host balance and to lay the foundation for chronic HBV infection.
