食管組織VEGF、HIF-1α、iNOS及ecNOS的表達導(dǎo)致食管靜脈曲張

【摘要】:為探討食管下段組織中血管內(nèi)皮生長因子(VEGF)、低氧誘導(dǎo)因子-1α(HIF-1α)、誘導(dǎo)型一氧化氮合酶(i NOS)及內(nèi)皮型一氧化氮合酶(ec NOS)的表達與門靜脈高壓癥下發(fā)生食管靜脈曲張之間的內(nèi)在關(guān)聯(lián),本研究選取Wistar大鼠90只,以門靜脈一步結(jié)扎法及結(jié)扎左腎上腺靜脈行食管靜脈曲張造模作為觀察組,并以僅行門靜脈主干分離,顯露左腎、左腎靜脈操作后的大鼠為對照組進行試驗觀察。研究發(fā)現(xiàn),建模后第7、14、28天,觀察組大鼠門靜脈壓力均顯著升高(p0.05);在14、28 d時,觀察組大鼠的HIF-1α、VEGF表達強度均明顯高于對照組(p0.05),且建模后各時間點i NOS、ec NOS表達也均明顯高于對照組(p0.05)。這表明VEGF、HIF-1α、i NOS及ec NOS的異常高表達,在食管靜脈曲張的形成過程中發(fā)揮了重要作用,可為臨床深入研究其發(fā)病機制提供理論依據(jù)。
[Abstract]:To investigate the relationship between the expression of vascular endothelial growth factor (VEGF), hypoxia inducible factor-1 偽 (HIF-1 偽), inducible nitric oxide synthase (iNOS) and endothelial nitric oxide synthase (Enos) and esophageal varices in patients with portal hypertension. In this study, 90 Wistar rats were selected as the observation group, the esophageal varices were made by one-step ligation of portal vein and ligation of left adrenal vein, and the left kidney was exposed by separating the main portal vein and exposing the left kidney. Rats treated with left renal vein were observed as control group. It was found that the portal vein pressure in the observation group was significantly higher than that in the control group at 1428 days after modeling, and the expression of HIF-1 偽 -VEGF in the observation group was significantly higher than that in the control group, and the expression of iNOS NOS was significantly higher than that in the control group at each time point after modeling. This suggests that the abnormal overexpression of NOS and EC NOS in VEGFU HIF-1 偽 may play an important role in the formation of esophageal varices, which may provide a theoretical basis for the further study of the pathogenesis of esophageal varices.
