轉(zhuǎn)化生長因子-β通路在腹腔間隔室綜合征大鼠模型腸黏膜損傷修復(fù)中的作用

【摘要】:目的探討轉(zhuǎn)化生長因子-β(TGF-β)信號通路在腹腔間隔室綜合征(ACS)大鼠模型腸黏膜損傷修復(fù)中的作用。方法 SD大鼠30只,隨機(jī)分為實驗組(ACS)組、對照組(sham組)。實驗組模型建立成功后再以相應(yīng)腹內(nèi)壓(20 mmHg)維持0.5、2、4、6 h 4個時間點隨機(jī)分為4個組,每組6只。每組實驗?zāi)┢?正中剖腹,找到回盲部,切取近端回腸組織,Q-PCR檢測TGF-β、cyclin D、CDK-2表達(dá)水平,Western blot檢測TGF-β、Smad2、磷酸化S-mad2(p-Smad2)蛋白表達(dá)水平;此外,如上相同分組共5只大鼠,處理同上,常規(guī)HE染色。結(jié)果 sham組回腸黏膜上皮細(xì)胞結(jié)構(gòu)完整,排列相對整齊;ACS組0.5 h腸黏膜損傷相對較輕,絨毛部分脫落;2 h時腸黏膜明顯水腫,腸黏膜上皮間隙增寬,黏膜上皮細(xì)胞變性及壞死;4 h和6 h腸絨毛完整性被破壞,絨毛頂端脫落,廣泛糜爛,中性粒細(xì)胞浸潤。ACS組0.5、2、4、6 h組TGF-β表達(dá)量較sham組表達(dá)降低(P0.05),cyclin D、CDK-2表達(dá)量較Sham組升高,差異有統(tǒng)計學(xué)意義(P0.05)。ACS組中TGF-β、p-Smad2蛋白表達(dá)較sham組降低,差異有統(tǒng)計學(xué)意義(P0.05),S-mad2蛋白表達(dá)在sham組和ACS組中差異無統(tǒng)計學(xué)意義(P0.05)。結(jié)論ACS形成后會造成腸黏膜損傷,腸黏膜損+傷后TGF-β信號通路啟動一系列分子反應(yīng),促進(jìn)腸黏膜損傷的修復(fù)。
[Abstract]:Objective to investigate the role of transforming growth factor- 尾 (TGF- 尾) signaling pathway in the repair of intestinal mucosal injury in rats with abdominal compartment syndrome (ACSS). Methods Thirty SD rats were randomly divided into experimental group and control group. After the establishment of the model, the experimental group was divided into 4 groups with 6 rats in each group. At the end of each experiment, the ileocecal part was found, and the expression level of TGF- 尾 cyclin DnCDK-2 in proximal ileum was detected by Q-PCR. The expression of TGF- 尾 -Smad2, phosphorylS-mad2p-Smad2) protein was detected by Western blot, and 5 rats were divided into the same group as above. Results the structure of ileal mucosal epithelial cells in sham group was intact, the intestinal mucosal injury was relatively mild in sham group at 0.5 h, and the intestinal mucosal mucosal edema was observed at 2 h after partial villi shedding, and the epithelial space of intestinal mucosa widened. The integrity of intestinal villi was destroyed at 4 h and 6 h after denaturation and necrosis of mucosal epithelial cells, the top of villi fell off, and there was extensive erosion. The expression of TGF- 尾 in the neutrophil infiltration group was significantly lower than that in the sham group. The expression of TGF- 尾 in the neutrophil infiltration group was significantly higher than that in the Sham group, and the expression of TGF- 尾 in the ACS group was significantly lower than that in the Sham group. The expression of TGF- 尾 p-Smad2 protein was significantly lower in ACS group than that in sham group. There was no significant difference in P0.05- Smad2 protein expression between sham group and ACS group. Conclusion the formation of ACS may lead to intestinal mucosal injury. TGF- 尾 signaling pathway after intestinal mucosal injury can initiate a series of molecular reactions and promote the repair of intestinal mucosal injury.
