外源性硫化氫對小鼠原代肝細胞內(nèi)脂質(zhì)自噬分解的影響

【摘要】:目的:研究硫化氫(H2S)供體GYY4137釋放的外源性H2S對小鼠原代肝細胞內(nèi)脂質(zhì)自噬分解的影響。方法:采用2步原位灌流法分離C57BL/6小鼠原代肝細胞并分為4組:用正常培養(yǎng)基培養(yǎng)正常組細胞;模型組用含1.2 mmol/L油酸(溶于10%的BSA)的培養(yǎng)基培養(yǎng)48 h;H2S組和炔丙基甘氨酸(PAG)組則用1.2 mmol/L油酸(溶于10%的BSA)的培養(yǎng)液培養(yǎng)48 h,然后換無血清無酚紅的RPMI-1640培養(yǎng)液(同時分別給予1 mmol/L GYY4137和200μmol/L PAG)處理6 h。收集各組細胞做LC3免疫熒光染色,拍攝熒光和相差圖片;Western blot檢測肝細胞中LC3-Ⅰ/Ⅱ的蛋白表達量;透射電鏡觀察肝細胞超微結(jié)構(gòu)。結(jié)果:和模型組相比,H2S組肝細胞內(nèi)LC3熒光顆粒和蛋白表達量增加,自噬溶酶體數(shù)量增加,細胞內(nèi)空泡增多。結(jié)論:外源性H2S可促進脂肪變性肝細胞內(nèi)脂質(zhì)自噬分解。
