乙型肝炎病毒影響肝癌HepG2細(xì)胞代謝的氣相色譜-質(zhì)譜研究

【摘要】:目的·研究乙型肝炎病毒(HBV)感染對(duì)肝癌HepG2細(xì)胞代謝的影響。方法·采用氣相色譜 質(zhì)譜聯(lián)用技術(shù)(GC-MS)檢測(cè)肝癌HepG2細(xì)胞和HepG2.2.15細(xì)胞(HepG2細(xì)胞的衍生細(xì)胞)的代謝物組成,分析其差異。結(jié)果·GC-MS分析發(fā)現(xiàn)HepG2及HepG2.2.15細(xì)胞的親水性提取物中主要含有34種代謝物,包括甘氨酸、丙氨酸、纈氨酸、亮氨酸、異亮氨酸、脯氨酸、絲氨酸、蘇氨酸、甲硫氨酸、半胱氨酸、胱氨酸、天冬氨酸、谷氨酸、焦谷氨酸、苯丙氨酸、酪氨酸、色氨酸、次黃嘌呤、尿嘧啶、肌醇、乳酸、丁二酸、亞油酸、亞麻酸、軟脂酸、硬脂酸、尿素和膽固醇等,覆蓋糖酵解、脂肪酸代謝、氨基酸代謝、嘌呤與嘧啶代謝等多條途徑。HepG2.2.15細(xì)胞中乳酸、亞麻酸、丙氨酸、半胱氨酸的含量顯著低于HepG2細(xì)胞,亮氨酸、異亮氨酸、纈氨酸、苯丙氨酸、甲硫氨酸、色氨酸、脯氨酸、酪氨酸、肌醇、尿嘧啶等代謝物含量顯著高于HepG2細(xì)胞。結(jié)論·HBV感染會(huì)導(dǎo)致HepG2細(xì)胞氨基酸代謝和脂類代謝改變,基于GC-MS的代謝組學(xué)技術(shù)能為HBV感染的代謝應(yīng)答機(jī)制提供補(bǔ)充信息。
[Abstract]:Objective to study the effect of hepatitis B virus (HBV) infection on the metabolism of hepatocellular carcinoma (HepG2) cells.Methods Gas chromatography-mass spectrometry (GC-MS) was used to detect the metabolites of hepatoma HepG2 cells and HepG2.2.15 cells derived from HepG2 cells.Results GC-MS analysis showed that the hydrophilic extracts of HepG2 and HepG2.2.15 cells mainly contained 34 metabolites, including glycine, alanine, valine, leucine, isoleucine, proline, serine, threonine, methionine, cysteine.Cystine, aspartic acid, glutamate, pyroglutamic acid, phenylalanine, tyrosine, tryptophan, Hypoxanthine, uracil, inositol, lactic acid, succinic acid, linoleic acid, linolenic acid, palmitic acid, stearic acid, urea and cholesterol, etc.,The contents of lactic acid, linolenic acid, alanine and cysteine in HepG2.2.15 cells were significantly lower than those in HepG2 cells, leucine, isoleucine and valine.The contents of phenylalanine, methionine, tryptophan, proline, tyrosine, inositol and uracil were significantly higher than those in HepG2 cells.Conclusion HBV infection may lead to the changes of amino acid and lipid metabolism in HepG2 cells. The metabolomics based on GC-MS can provide supplementary information for the metabolic response mechanism of HBV infection.
