非酒精性脂肪肝大鼠腸道黏膜屏障功能的實驗研究

【摘要】:目的腸道黏膜屏障功能紊亂與非酒精性脂肪肝(non-alcohol fatty liver disease,NAFLD)的發(fā)生關系密切,文中旨在通過動物模型的方法探討NAFLD大鼠腸道黏膜屏障功能。方法 SD大鼠16只按隨機數(shù)字表法分為正常組和NAFLD組,每組8只。正常組由普通飼料喂養(yǎng),每100克普通飼料提供326 k Cal熱量。NAFLD組大鼠由高脂飼料喂養(yǎng),每100克高脂飼料提供453 k Cal熱量建立NAFLD模型。采用HE染色檢測肝病理,Elisa法和鱟試劑法分別檢測腫瘤壞死因子-α(tumor necrosis factor-α,TNF-α)、白細胞介素-1(Interleukin-1,IL-1)、內(nèi)毒素,實時定量PCR法檢測小腸閉鎖小帶蛋白ZO-1和連接蛋白Occluding的表達。結果 NAFLD組大鼠肝呈現(xiàn)典型的脂質沉積,血漿內(nèi)毒素和血清TNF-α、IL-1較正常組顯著增高,小腸ZO-1和Occluding表達較正常組顯著降低(P0.05)。結論 NAFLD大鼠存在腸道黏膜屏障功能下降。
[Abstract]:Objective to investigate the relationship between intestinal mucosal barrier dysfunction and the occurrence of non-alcoholic fatty liver disease (NAFLD) in NAFLD rats.Methods Sixteen SD rats were randomly divided into normal group and NAFLD group with 8 rats in each group.The normal group was fed with normal diet, and the rats of NAFLD group were fed with high fat diet and every 100g high fat diet with 453k Cal calorie per 100g normal diet to establish NAFLD model.The expression of small intestinal atresia band protein (ZO-1) and connectin Occluding (Occluding) were detected by HE staining and Limulus lysate assay respectively. The expression of TNF- 偽, Interleukin-1 and endotoxin in small intestinal atresia band protein (ZO-1) and connectin Occluding (Occluding) were detected by real-time quantitative PCR.Results the liver of NAFLD group showed typical lipid deposition, plasma endotoxin and serum TNF- 偽 IL-1 were significantly higher than those in normal group, and the expression of ZO-1 and Occluding in small intestine was significantly lower than that in normal group.Conclusion the intestinal mucosal barrier function is decreased in NAFLD rats.
