神經(jīng)生長因子對(duì)四氯化碳誘導(dǎo)的小鼠肝纖維化的影響

【摘要】:目的觀察神經(jīng)生長因子(NGF)對(duì)四氯化碳(CCL4)誘導(dǎo)的小鼠肝纖維化的影響。方法將30只雌性昆明小鼠分成3組:纖維化模型組(A組)、NGF干預(yù)組(B組)和生理鹽水對(duì)照組(C組)。8周時(shí)采集標(biāo)本,用全自動(dòng)生化分析儀檢測丙氨酸氨基轉(zhuǎn)移酶(ALT)、天冬氨酸氨基轉(zhuǎn)移酶(AST)、總膽紅素(TBIL)、清蛋白(ALB);放射免疫法檢測肝纖維化指標(biāo):透明質(zhì)酸(HA)、層粘連蛋白(LN)、Ⅲ型前膠原(PCⅢ)。采用Ishaki評(píng)分系統(tǒng)對(duì)小鼠肝組織炎癥活動(dòng)度及纖維化程度進(jìn)行評(píng)分。結(jié)果A、B組ALT、AST、HA、LN均明顯高于C組(F=111.45,658.80,157.43,167.99;P0.05);B組ALT、AST、LN均明顯低于A組(P0.05)。HE染色、網(wǎng)狀纖維染色及Masson染色顯示,A組肝組織炎癥及纖維化程度最明顯,B組肝組織炎癥較A組顯著減輕,未形成纖維間隔,纖維組織細(xì)短,C組肝組織未見明顯炎癥細(xì)胞浸潤及纖維形成。A組肝臟炎癥分級(jí)和纖維化分期評(píng)分較B、C組高,且3組炎癥活動(dòng)度得分、纖維化得分差異有統(tǒng)計(jì)學(xué)意義(均P0.05)。結(jié)論 NGF可以阻斷CCL4誘導(dǎo)的小鼠肝纖維化,減輕肝組織炎癥。
