谷氨酰胺對(duì)梗阻性黃疸大鼠小腸黏膜組織Akt活性的影響

【摘要】:目的觀察梗阻性黃疸大鼠小腸組織中蛋白激酶B(Akt)活性,探討谷氨酰胺腸保護(hù)的作用機(jī)制。方法將64只SD大鼠隨機(jī)分為4組各16只,模型組、甘氨酸組、谷氨酰胺組行梗阻性黃疸造模,假手術(shù)組游離上段膽總管后關(guān)腹;造模后1 d,甘氨酸組、谷氨酰胺組均給予同等熱量的腸內(nèi)營(yíng)養(yǎng)劑灌胃,同時(shí)分別給予甘氨酸、谷氨酰胺1 g/(kg·d),模型組、假手術(shù)組給予等量生理鹽水灌胃。各組分別于術(shù)后第3、7天處死大鼠8只,采用Western blotting法檢測(cè)小腸組織Akt總蛋白及p-Akt,實(shí)時(shí)PCR檢測(cè)腸組織Akt mRNA。結(jié)果與假手術(shù)組比較,模型組、甘氨酸組、谷氨酰胺組術(shù)后第3、7天小腸組織中Akt mRNA及p-Akt蛋白增加,且谷氨酰胺組高于模型組、甘氨酸組,P均0.05;模型組、甘氨酸組間比較,P均0.05。各組小腸組織Akt總蛋白比較,P均0.05。結(jié)論谷氨酰胺通過促進(jìn)Akt信號(hào)通路的活化,發(fā)揮對(duì)小腸上皮細(xì)胞的保護(hù)作用。
[Abstract]:Objective to observe the activity of protein kinase (PKB) Akt in small intestine of rats with obstructive jaundice and to explore the protective mechanism of glutamine. Methods Sixty-four SD rats were randomly divided into 4 groups, 16 rats in each group, model group and glycine group. The rats in the glutamine group were treated with obstructive jaundice, and in the sham operation group, the upper common bile duct was closed, and the glycine group and the glutamine group were given the same caloric enteral nutrition at the same time as glycine. The rats in the model group and sham operation group were given the same amount of normal saline intragastrically. The rats in each group were killed on the 3rd day after operation, and 8 rats were killed on the 7th day after operation. Total Akt protein and p-Akt in small intestine were detected by Western blotting method, and Akt mRNA in intestinal tissue were detected by real-time PCR. Results compared with sham operation group, Akt mRNA and p-Akt protein in small intestine were increased in model group, glycine group and glutamine group on the 3rd day after operation. Glutamine group was higher than model group (P 0.05), model group (P 0.05), glycine group (P 0.05) and small intestine Akt protein (P 0.05). Conclusion glutamine can promote the activation of Akt signal pathway. To play a protective role on small intestinal epithelial cells.
