飽和脂肪酸與單不飽和脂肪酸對HepG2細(xì)胞自噬和凋亡的影響

【摘要】:目的比較飽和脂肪酸棕櫚酸(palmitic acid,PA)和單不飽和脂肪酸油酸(oleic acid,OA)對HepG2細(xì)胞凋亡和自噬的影響。方法 PA和OA處理HepG2細(xì)胞,CCK-8檢測脂肪酸的細(xì)胞毒性,caspase-3活性測定檢測脂肪酸對細(xì)胞凋亡的影響,油紅O染色檢測細(xì)胞內(nèi)脂質(zhì)沉積,酶法檢測細(xì)胞內(nèi)甘油三酯的含量,Western blot檢測自噬相關(guān)蛋白LC3-Ⅱ和凋亡相關(guān)蛋白caspase-3的表達(dá),實(shí)時熒光定量PCR檢測相關(guān)基因表達(dá)。結(jié)果 PA和OA都均同時誘導(dǎo)HepG2細(xì)胞凋亡與自噬,但PA的影響明顯強(qiáng)于OA;PA誘導(dǎo)HepG2細(xì)胞自噬相關(guān)基因LC3B和VPS38表達(dá)、促進(jìn)脂肪酸β-氧化的PPARα基因mRNA水平升高,但對細(xì)胞內(nèi)脂肪沉積沒有明顯作用;而OA則導(dǎo)致HepG2細(xì)胞內(nèi)甘油三酯的水平明顯增加,但對上述基因表達(dá)沒有明顯影響。結(jié)論飽和脂肪酸PA和不飽和脂肪酸OA均能引起HepG2細(xì)胞凋亡和自噬,但PA的作用強(qiáng)于OA。
[Abstract]:Objective to compare the effects of palmitic acido) and monounsaturated fatty acid oleic acidOAA on apoptosis and autophagy of HepG2 cells. Methods HepG2 cells treated with PA and OA were treated with CCK-8 to detect the cytotoxicity of fatty acids and the activity of caspase-3 to detect fat.
