【摘要】：目的:探討含目的基因趨化因子生長(zhǎng)調(diào)節(jié)基因1(CXCL-1)慢病毒表達(dá)系統(tǒng)的構(gòu)建及CXCL-1對(duì)7702正常肝細(xì)胞生長(zhǎng)、增殖、遷移生物學(xué)功能的影響。方法:構(gòu)建含目的基因CXCL-1的慢病毒表達(dá)系統(tǒng),建立攜帶GFP熒光標(biāo)記并過(guò)度表達(dá)CXCL-1的7702正常肝細(xì)胞系,Western blot方法檢測(cè)CXCL-1在轉(zhuǎn)染細(xì)胞中的表達(dá),細(xì)胞單克隆形成實(shí)驗(yàn),MTT實(shí)驗(yàn),Transwell實(shí)驗(yàn)檢測(cè)7702細(xì)胞表達(dá)CXCL-1后生長(zhǎng)、增殖和遷移能力的變化情況。結(jié)果:1成功構(gòu)建目的基因CXCL-1慢病毒表達(dá)系統(tǒng),病毒滴度檢測(cè)結(jié)果為2.00E+8TU/mL;2轉(zhuǎn)染48h后經(jīng)流式細(xì)胞儀檢測(cè),轉(zhuǎn)染率達(dá)94.5%;3CXCL-1/7702組克隆形成率顯著高于其余兩對(duì)照組(P0.05);4MTT比色法結(jié)果顯示:CXCL-1/7702組分別與空載GFP/7702組及空白對(duì)照7702組相比較,生長(zhǎng)速度明顯較快(P0.05);5Transwell實(shí)驗(yàn)顯示:CXCL-1/7702組、GFP/7702組、7702組肝細(xì)胞系均無(wú)遷移能力。結(jié)論:成功構(gòu)建含目的基因CXCL-1的慢病毒表達(dá)系統(tǒng),CXCL-1對(duì)正常肝細(xì)胞具有明顯的促細(xì)胞生長(zhǎng)與增殖功能,但無(wú)促進(jìn)正常肝細(xì)胞遷移的功能。
[Abstract]:Objective: to investigate the construction of lentivirus expression system containing chemokine growth regulator gene 1 (CXCL-1) and the effect of CXCL-1 on the growth, proliferation and migration of 7702 normal hepatocytes. Methods: a lentivirus expression system containing the target gene CXCL-1 was constructed. A normal liver cell line 7702 carrying GFP fluorescent labeling and overexpression of CXCL-1 was established to detect the expression of CXCL-1 in transfected cells by Western blot. Results the target gene CXCL-1 lentivirus expression system was successfully constructed. The results showed that the virus titer was 2.00E8TU / mLn-2 transfection for 48h, then detected by flow cytometry. The clone formation rate of group 1 / 7702 was significantly higher than that of the other two control groups (P0.05CL-1 / 7702). The results showed that the colony formation rate of the control group was significantly higher than that of the control group (7702) and the control group (7702), compared with the control group (7702) and the control group (7702). The growth rate was significantly faster than that of P0.05 / 5Transwell experiment. The results showed that the liver cell lines of GFP / 7702 group had no migration ability. Conclusion: the lentivirus expression system (CXCL-1) containing the target gene CXCL-1 can promote the growth and proliferation of normal hepatocytes, and CXCL-1 can promote the growth and proliferation of normal hepatocytes. However, it had no function to promote the migration of normal hepatocytes.
