熊去氧膽酸改善膽汁淤積癥幼鼠肝功能和抗肝纖維化的作用

【摘要】:本研究試圖通過制造幼鼠膽汁淤積癥模型進(jìn)而探討熊去氧膽酸(ursodeoxycholic acid,UDCA)對(duì)膽汁淤積癥幼鼠肝功能及肝纖維化指標(biāo)的作用及影響。我們采用α-萘異硫氰酸酯(α-naphthyl isothiocyanate,ANIT)灌胃誘發(fā)幼鼠膽汁淤積癥模型,檢測(cè)經(jīng)UDCA治療后對(duì)大鼠肝功能各項(xiàng)生化指標(biāo)、血清肝纖四項(xiàng)、肝組織免疫組化影響。結(jié)果提示造模組(ANIT Model)除膽堿酯酶(cholinesterase,CHE)外,肝功能各項(xiàng)指標(biāo)均明顯高于空白對(duì)照組(blank control)(p0.01),表明造模成功;試驗(yàn)顯示:γ-谷酰轉(zhuǎn)肽酶(γ-glutamyl transpep tidase,γ-GT)、膽汁酸(total bile acids,TBA)在熊去氧膽酸治療組(UDCA Treatment)中數(shù)值,明顯較ANIT Model組降低,有統(tǒng)計(jì)學(xué)差異(p0.05),而丙氨酸氨基轉(zhuǎn)移酶(alanine aminotransferase,ALT)、天門冬氨酸氨基轉(zhuǎn)移酶(aspartate aminotransferase,AST)、總膽紅素(total bilirubin,TBIL)、直接膽紅素(direct bilirubin,DBIL)、間接膽紅素(indirect bilirubin,IBIL)數(shù)值,相比ANIT Model組,無統(tǒng)計(jì)學(xué)差異;UDCA Treatment組血清層粘連蛋白(laminin,LN)、Ⅳ型膠原(collagenⅣ,cⅣ)明顯低于ANIT Model組(p0.01),而透明質(zhì)酸(hyaluronic acid,HA)、Ⅲ型前膠原(procollagenⅢ,PCⅢ)與ANIT Model組相比均無統(tǒng)計(jì)學(xué)差異;UDCA Treatment組肝組織LN、cⅣ均明顯低于ANIT Model組(p0.01),有統(tǒng)計(jì)學(xué)意義。因此,通過本研究證明了UDCA有可能通過降低血清γ-GT、TBA改善肝功能,通過降低血清及肝組織中LN、cⅣ而起到抗肝纖維化作用。
[Abstract]:The purpose of this study was to investigate the effects of ursodeoxycholic acid (UDCA) on liver function and liver fibrosis in young rats with cholestasis by making a model of cholestasis in young rats. We used 偽 -naphthyl isothiocyanate (偽 -naphthyl isothiocyanate) to induce intragastric perfusion of liver function and liver fibrosis indexes in young rats with cholestasis. The model of cholestasis in young rats, The biochemical indexes of liver function, four items of serum hepatic fiber, and the effect of immunohistochemistry on liver function of rats treated with UDCA were detected. The results suggested that the model group was not only cholinesterase cholinesterase (che). All the indexes of liver function were significantly higher than those of blank control group (P 0.01), which indicated that the model was successful, and the results showed that the value of 緯 -glutamyl transpep tidase (緯 -GTase, total bile acidsTBAs) in ursodeoxycholic acid treatment group was significantly lower than that in ANIT Model group. There was a statistical difference between the two groups, and alanine aminotransferase (alt), aspartate aminotransferase (AST), total bilirubin (TBI), direct bilirubin (DBIL), indirect bilirubin (IBIL), and aspartate aminotransferase (aspartate aminotransferase) were found in the ANIT Model group.
