白介素-7對(duì)CCL4誘導(dǎo)的小鼠肝纖維化作用及影響因素

【摘要】:目的探討白介素-7對(duì)CCL4誘導(dǎo)的小鼠肝纖維化作用及影響因素。方法用四氯化碳(CCl4)誘導(dǎo)形成小鼠肝纖維化模型。將雌性balb/c小鼠36只隨機(jī)分成3組:A組(生理鹽水對(duì)照組),B組(纖維化模型組),C組(IL-7干預(yù)組),每組12只。每個(gè)組隨機(jī)再分為兩個(gè)亞組,分別記為A1、A2,B1、B2及C1、C2(1表示2周,2表示4周),每亞組6只。A1、B1、C1組及A2、B2、C2組分別于實(shí)驗(yàn)2周和4周后取血并處死取肝臟,用ELASA法檢測(cè)各組小鼠血清中ALT、AST濃度,肉眼及組織病理形態(tài)學(xué)法(常規(guī)HE染色、masson染色及網(wǎng)狀纖維染色)觀察肝組織標(biāo)本,并采用Ishak半定量評(píng)分系統(tǒng)對(duì)小鼠肝組織炎癥活動(dòng)度及纖維化程度進(jìn)行評(píng)分。用SPSS18.0統(tǒng)計(jì)軟件對(duì)實(shí)驗(yàn)數(shù)據(jù)進(jìn)行分析處理。結(jié)果成功建立小鼠肝纖維化模型,表現(xiàn)為匯管區(qū)及周圍纖維擴(kuò)大,有芒狀纖維和小葉纖維間隔形成。血生化檢測(cè):同時(shí)期相比,B、C組ALT及AST均顯著高于A組(P0.05),C組ALT及AST均顯著低于B組(P0.05);同組內(nèi)不同時(shí)間亞組比較:B2組AST顯著高于B1組(P0.05),C2組ALT及AST均顯著低于C1組(P0.05)。HE染色、Masson染色和網(wǎng)狀纖維染色均顯示:同時(shí)期相比,B組小鼠炎癥及纖維化程度均最嚴(yán)重,C組較B組輕;亞組間比較:B2組炎癥及纖維化程度均較B1組重,而C2組較C1組輕;Ishak炎癥活動(dòng)度及肝纖維化程度評(píng)分及統(tǒng)計(jì)學(xué)分析示:同時(shí)期三組相比,B組炎癥及纖維化評(píng)分均最高,三組之間差異顯著(P0.05),同組內(nèi)亞組比較:B2炎癥及纖維化評(píng)分均顯著高于B1組(P0.05);C2組炎癥評(píng)分及纖維化評(píng)分均低于C1組,炎癥評(píng)分差異顯著(P0.05),纖維化評(píng)分差異不明顯(P0.05)。結(jié)論白介素-7能抑制CCl4誘導(dǎo)的小鼠肝纖維化,減輕肝臟炎癥,作用效果與干預(yù)時(shí)間相關(guān)。
[Abstract]:Objective to investigate the effect of interleukin-7 (IL-7) on CCL4 induced liver fibrosis in mice and its influencing factors. 36 female balb/c mice were randomly divided into 3 groups: group A (normal saline control group). Group B (Fibrosis model group) treated with IL-7 group (12 rats in each group). Each group was randomly divided into two subgroups, which were marked as A1A2OB1B1B2 and C1C2O1 respectively for 2 weeks. (2) for 4 weeks, 6 rats in each subgroup. The blood was taken from the blood and the liver was killed in the group A _ 2 B _ 2C _ 2 and A _ (1) B _ (1) C _ (1) and A _ (2) B _ (2) C _ 2 after 2 and 4 weeks, respectively. The concentration of alt AST in serum, histopathology (routine HE staining, mass on staining and reticular fiber staining) were detected by ELASA method. Ishak semi-quantitative scoring system was used to evaluate the degree of inflammatory activity and fibrosis in liver tissue of mice. The experimental data were analyzed and processed by SPSS18.0 software. Results the mice were successfully established. Liver fibrosis model. The blood biochemical test showed that the ALT and AST of group C were significantly higher than that of group A (P 0.05). ALT and AST in group C were significantly lower than those in group B (P 0.05). The AST of group B 2 was significantly higher than that of group B1 (P 0.05) and AST of group C 2 was significantly lower than that of group C 1 (P 0.05). Masson staining and reticular fiber staining showed that the severity of inflammation and fibrosis in group B was lighter than that in group B at the same time. The degree of inflammation and fibrosis in group B _ 2 was heavier than that in group B1, while that in group C _ 2 was lighter than that in group C _ 1. Ishak inflammatory activity and liver fibrosis degree score and statistical analysis showed that: the same period of the three groups compared with the B group inflammation and fibrosis score were the highest, the difference between the three groups was significant (P 0.05). The score of inflammation and fibrosis in group B 2 was significantly higher than that in group B 1 (P 0.05). The inflammation score and fibrosis score of C2 group were lower than that of C1 group, and the difference of inflammation score was significant (P0.05). Conclusion Interleukin-7 can inhibit CCl4 induced liver fibrosis and alleviate liver inflammation in mice. The effect is related to the time of intervention.
