視網(wǎng)膜色素上皮（retinal pigment epithelium,RPE）細(xì)胞和光感受器細(xì)胞的丟失、變性或壞死均可導(dǎo)致一系列視網(wǎng)膜變性疾病,包括視網(wǎng)膜色素變性（retinitis pigmentosa,RP）,老年性黃斑變性（age-related macular degeneration,AMD）,先天性黑朦（Lebers congenital amaurosis,LCA）和視錐-視桿細(xì)胞營養(yǎng)不良（cone-rod dystrophy,CORD）等疾病。目前,國內(nèi)外尚無該類嚴(yán)重性致盲性疾病的有效治療手段。關(guān)于干細(xì)胞替代性治療視網(wǎng)膜變性疾病的可行性和有效性,近年來成為該領(lǐng)域的研究重點(diǎn)和熱點(diǎn)。胚胎干細(xì)胞（embryonic stem cell,ESC）具有分化的多潛能性,來源穩(wěn)定且能在體外大量擴(kuò)增,使其成為治療視網(wǎng)膜變性疾病的一個(gè)研究熱點(diǎn),為視網(wǎng)膜變性疾病提供一種全新的治療模式,為有效治療嚴(yán)重致盲性眼部疾病帶來了希望。研究發(fā)現(xiàn),在一定的培養(yǎng)條件下,ESCs能被誘導(dǎo)分化為神經(jīng)細(xì)胞、胰島細(xì)胞、造血細(xì)胞、心肌細(xì)胞、骨細(xì)胞和肝細(xì)胞等,涵括了內(nèi)、中、外三個(gè)胚層的所有細(xì)胞和組織,包括RPE細(xì)胞和...
