抑制單羧酸轉(zhuǎn)運蛋白1可增強鼻咽癌細胞HNE1/DDP對順鉑誘導(dǎo)凋亡的敏感性

目的探討siRNA沉默單羧酸轉(zhuǎn)運蛋白1(MCT1)增強鼻咽癌細胞HNE1/DDP對順鉑誘導(dǎo)凋亡的敏感性及其可能機制。方法 Western blot檢測HNE1和HNE1/DDP細胞中MCT1的表達及siRNA轉(zhuǎn)染沉默MCT1在HNE1/DDP細胞中MCT1的表達;MTT法檢測不同濃度順鉑和MCT1 siRNA聯(lián)合順鉑對HNE1/DDP細胞的增殖抑制作用;PI單染流式細胞術(shù)檢測細胞凋亡;線粒體膜電位檢測試劑盒(JC-1)檢測細胞線粒體膜電位;Western blot檢測Mcl-1、Bak、Bcl-2、Bax蛋白的表達。結(jié)果 MCT1在HNE1/DDP細胞中高表達,抑制MCT1的表達可增加HNE1/DDP細胞對順鉑的敏感性(P<0.05),部分逆轉(zhuǎn)鼻咽癌細胞對順鉑的耐藥。此外,抑制MCT1的表達可增強HNE1/DDP細胞對順鉑誘導(dǎo)凋亡的作用,MCT1 siRNA與8μmol/L順鉑聯(lián)合作用于HNE1/DDP細胞24 h的凋亡率為(51.23±2.86)%,較單用MCT1 siRNA、順鉑的凋亡率明顯升高(P<0.05)。同時使細胞線粒體膜電位降低,下調(diào)Mcl-1、Bcl-2的表...
