miR-126通過阻礙VEGFA和PIK3R2抑制高糖下RF/6A細(xì)胞的遷移和管腔形成

第一部分高糖條件下RF/6A細(xì)胞中miR-126及其相關(guān)因子的表達(dá)情況[目的]檢測(cè)RF/6A細(xì)胞在標(biāo)準(zhǔn)培養(yǎng)液,高糖培養(yǎng)液和高滲培養(yǎng)液三個(gè)培養(yǎng)條件下miR-126,VEGFA和PIK3R2的表達(dá)情況。[方法]利用q-PCR和western-blot檢測(cè)標(biāo)準(zhǔn)培養(yǎng)液(NG,D-葡萄糖:2mrnol/L),高糖培養(yǎng)液(HG,D-葡萄糖:10mmol/L)和高滲培養(yǎng)液(OSM,NG,D-葡萄糖:2mmol/L+L-葡萄糖:8mmol/L)三個(gè)不同糖濃度條件下培養(yǎng)的RF/6A 細(xì)胞中 miR-126、VEGFA 和 PIK3R2 的表達(dá)。[結(jié)果]1、在NG組,miR-126相對(duì)表達(dá)量為(1.00±0.00);在HG組,miR-126相對(duì)表達(dá)量為(0.70±0.07);在OSM組,miR-126相對(duì)表達(dá)量為(0.97±0.02)。比較各組結(jié)果:與NG組和OSM組比,HG組RF/6A細(xì)胞的miR-126表達(dá)均降低(P<0.05);與NG組比,OSM組RF/6A細(xì)胞的miR-126表達(dá)變化無統(tǒng)計(jì)學(xué)差異(P>0.05)。2、在 NG 組,VEGFAmR]NA 相對(duì)表達(dá)量為(1.00±0.00)...
