經(jīng)鼻滴入抗白介素5多克隆抗體治療大鼠變應(yīng)性鼻炎的機(jī)理
[Abstract]:Objective: to investigate the mechanism of anti-interleukin-5 polyclonal antibody (IL-5) in the treatment of allergic rhinitis in rats with (AR). Methods: the (AR) model of allergic rhinitis in rats was established by using ovalbumin (OVA) and aluminum hydroxide. They were divided into three groups: model group (group B), corticosteroid group (group C) and anti-IL-5 antibody group (group D) with 6 rats in each group. After the establishment of the model, fluticasone propionate was injected into nasal cavity of rats in group C for 7 days, while rats in group D were injected with IL-5 antibody 30L every day for one week, and rats in group B were killed. The percentage of eosinophils in nasal lavage fluid and bone marrow cell suspension were measured. Histopathological changes of nasal mucosa and the expression of GATA-3 were observed by mucosal sections. Results: the percentage of eosinophils in nasal lavage fluid and bone marrow lavage fluid in group D was significantly lower than that in group B (P 0.01? P 0.01). Pathological sections showed that the nasal mucosa of group B was thickened, accompanied by a large number of inflammatory cell infiltration, mainly eosinophils, mucosal congestion, edema, sparse cilia. The changes of inflammatory symptoms in group D were significantly less than those in group B; The results of immunohistochemistry showed that the expression of GATA-3 in nasal mucosa of group B was increased while that of group D was decreased. The average optical density of nasal mucosa of group D was significantly lower than that of group B (P 0.01). Conclusions: 1. Anti-IL-5 antibody has obvious therapeutic effect on allergic rhinitis in rats. The therapeutic effect of anti-IL-5 antibody on allergic rhinitis in rats was shown to protect the ciliated structure of the nasal mucosa and decrease the infiltration or distribution of eosinophils in the nasal and bone marrow at the cellular level. 3. The decrease of eosinophils infiltration in nasal mucosa by anti-IL-5 antibody is related to its significant inhibition of the protein expression of GATA-3 transcription factor in nasal mucosa of rats.
