光催化納米二氧化鈦對(duì)人鼻咽癌裸鼠模型的治療作用研究

【摘要】:目的:研究光催化納米二氧化鈦(TiO_2)對(duì)人鼻咽癌裸鼠移植瘤模型的治療效果,初步探討其抑癌機(jī)理,評(píng)估其對(duì)機(jī)體的影響,為臨床研究應(yīng)用納米TiO_2光動(dòng)力療法治療鼻咽癌提供理論及實(shí)驗(yàn)依據(jù)。 方法:建立荷人鼻咽癌裸鼠移植瘤模型,隨機(jī)分成6組,包括實(shí)驗(yàn)組:梯度濃度的納米TiO_2+紫外光照射組;陰性對(duì)照組:?jiǎn)渭兗{米TiO_2組與單純紫外光照射組;空白對(duì)照組。其中納米TiO_2懸液采用瘤內(nèi)注射,經(jīng)波長(zhǎng)范圍330-400nm的紫外光處理后24h,,取各組瘤組織及肝、腎HE染色做病理分析;透射電鏡觀察腫瘤細(xì)胞超微結(jié)構(gòu)的變化及凋亡情況;采用免疫組化檢測(cè)瘤組織caspase-3的表達(dá),TUNEL法檢測(cè)細(xì)胞凋亡情況,并分析兩者相關(guān)性。 結(jié)果:1、病理學(xué)分析,納米TiO_2光催化治療后移植瘤組織的局部炎癥反應(yīng)明顯(Ⅱ、Ⅲ級(jí)),伴隨炎癥反應(yīng)的是局部壞死灶形成;而陰性及空白對(duì)照組無明顯炎癥反應(yīng)(Ⅰ級(jí))。高倍鏡觀察,各組肝、腎組織均未見明顯急性炎癥反應(yīng)或組織損傷表現(xiàn)。 2、透射電鏡下觀察,實(shí)驗(yàn)組可見納米TiO_2顆粒廣泛進(jìn)入胞漿胞核,腫瘤細(xì)胞多有凋亡的形態(tài)學(xué)改變。 3、實(shí)驗(yàn)組caspase-3的陽(yáng)性表達(dá)及TUNEL標(biāo)記的陽(yáng)性細(xì)胞數(shù)顯著多于對(duì)照組(P0.01),且陽(yáng)性表達(dá)隨TiO_2濃度遞增而增強(qiáng)(P0.01)。 結(jié)論:1、光催化納米TiO_2可通過提高移植瘤組織的caspase-3表達(dá)活性來誘導(dǎo)細(xì)胞凋亡,抑制腫瘤生長(zhǎng),且效果在一定范圍內(nèi)呈TiO_2濃度依賴性。 2、光催化納米TiO_2通過誘導(dǎo)腫瘤細(xì)胞凋亡和介導(dǎo)炎癥反應(yīng)是其發(fā)揮抑 癌作用的可能機(jī)制。 3、對(duì)機(jī)體無明顯毒副作用,這為光催化納米TiO_2技術(shù)有望應(yīng)用于臨床治療鼻咽癌提供實(shí)驗(yàn)保障。
[Abstract]:Objective: To study the effect of photocatalytic nano-titanium dioxide (TiO _ 2) on the tumor model of human nasopharyngeal carcinoma (NPC) in nude mice. Methods: The model of nude mice with human nasopharyngeal carcinoma (NPC) was established and divided into 6 groups, including the experimental group, the nano-TiO _ 2 + ultraviolet irradiation group, the negative control group, the pure nano-TiO _ 2 group and the pure ultraviolet irradiation group, and the blank control group. The changes of the ultrastructure of the tumor cells and the apoptosis of the tumor cells were observed by transmission electron microscope, and the expression of caspase-3 was detected by immunohistochemistry. The apoptosis of the cells was detected by the TUNEL method, and their correlation was analyzed. Results:1.
