原花青素對人眼小梁細胞抗氧化損傷的實驗研究

【摘要】:目的: 研究原花青素(Procyanidins,PC)對過氧化氫(Hydrogen peroxide)處理的人眼小梁細胞(Human Trabecular Meshwork Cells,HTMC)抗氧化損傷的保護作用,為青光眼的臨床治療提供實驗依據(jù)。 方法: 將正常人眼小梁細胞進行細胞傳代后隨機分為5組。未處理組:正常培養(yǎng)的HTMC,對照組:正常培養(yǎng)的HTMC+過氧化氫;實驗組:正常培養(yǎng)的HTMC+過氧化氫+PC(PC濃度分別為0.02mg/ml、0.05mg/ml、0.10mg/ml)。應(yīng)用實時熒光定量PCR方法檢測線粒體復合物I mRNA(complex Ⅰ mRNA)的表達,流式細胞術(shù)檢測細胞凋亡率,Western blot檢測細胞色素C(cytochrome C,CytC)的釋放。 結(jié)果: 1.與對照組相比,各實驗組線粒體complex Ⅰ mRNA表達均增加,CytC釋放均減少,細胞凋亡率均降低,差異均有統(tǒng)計學意義(P0.01)。2.不同濃度PC組間隨PC濃度增加complex Ⅰ mRNA表達增加,CytC釋放減少,細胞凋亡率降低,各濃度組間差異有統(tǒng)計學意義(P0.01)。3.與未處理組相比,各PC組細胞凋亡率差異均有統(tǒng)計學意義(P0.01),,0.02mg/ml和0.05mg/mlPC組complexI mRNA表達、CytC釋放差異均有統(tǒng)計學意義(P0.01),而0.10mg/mlPC組差異均無統(tǒng)計學意義(P0.05)。 結(jié)論: 外源性PC可以增加氧化損傷人眼小梁細胞線粒體complex Ⅰ mRNA的表達,減少CytC的釋放,降低細胞凋亡率,有較強的抗氧化作用,并在一定濃度范圍內(nèi)隨著濃度的增加其抗氧化作用逐漸增強,PC在青光眼的臨床治療中的作用值得進一步研究。
