大鼠局灶性腦缺血再灌注后聽覺中樞損傷的研究
[Abstract]:Objective to investigate the changes of auditory center and the possible mechanism of hearing impairment after focal cerebral ischemia reperfusion injury in rats. Methods 60 healthy white male SD rats were randomly divided into sham operation group and ischemia reperfusion group with 30 rats in each group. The middle cerebral artery embolization model of rats was established by thread embolization. The sham-operated group only separated cervical blood vessels and did not insert thread embolism. Cerebral ischemia for 60 min and reperfusion for 24 h. Auditory brainstem reaction was detected before and 24 hours after operation. Nerve function score, water content of brain tissue were measured by dry and wet weight method, cerebral infarction volume was measured by TTC method, and the integrity of blood-brain barrier was evaluated by the exudation of Evans blue. The apoptosis of nerve cells was observed by end labeling method, and the expression of MMP-9,Clau-din-5,Occludin,PSD-95,CX-43 and Na-偽 protein was measured by. Western blotting method. Results compared with the sham-operation group, the neurological function score and cerebral water content in the ischemia-reperfusion group increased, the volume of cerebral infarction increased, the threshold of ABR reaction increased, the apoptotic index increased, the expression of MMP-9,CX-43 protein increased, and the expression of Claudin-5,Occludin, increased. The expression of PSD-95,Na-偽 protein decreased significantly. The difference between the two groups was statistically significant. Conclusion the changes of auditory center and hearing loss after focal cerebral ischemia-reperfusion injury may be related to the activation of MMPs, the destruction of blood-brain barrier, the activation of gap junction channel, the abnormal synaptic function and the destruction of ion channel.
