長期注射水楊酸鈉大鼠下丘EphA4的表達(dá)

【摘要】:目的通過觀察長期水楊酸鈉作用后大鼠ABR及下丘EphA4mRNA表達(dá)變化,探討下丘EphA4表達(dá)在水楊酸鈉耳毒性中的作用。方法將30只健康成年雄性SD大鼠隨機(jī)分為5組,每組6只:正常對照組(不予任何處理),肌注7天組(肌肉注射水楊酸鈉175mg/kg,每天2次,間隔8小時(shí),連續(xù)7天),肌注14天組(水楊酸鈉用法同前,持續(xù)14天),恢復(fù)14天組(肌注水楊酸鈉14天后,停藥恢復(fù)14天),恢復(fù)28天組(肌注水楊酸鈉14天后,停藥恢復(fù)28天)。對各組大鼠分別于相應(yīng)時(shí)間點(diǎn)進(jìn)行ABR檢測后處死并迅速分離下丘,采用實(shí)時(shí)熒光定量PCR(real-time PCR)的方法檢測各組大鼠下丘中EphA4mRNA表達(dá)。結(jié)果 1肌注7、14天組大鼠的ABR反應(yīng)閾分別為38.33±3.73、44.16±1.86dB SPL,較對照組(30.83±1.86dB SPL)明顯升高(P0.05);恢復(fù)28天組大鼠的ABR反應(yīng)閾(32.50±2.50dB SPL)與對照組(30.83±1.86dB SPL)相比,差異無明顯統(tǒng)計(jì)學(xué)意義(P0.05);2肌注7天組和恢復(fù)14天組大鼠下丘EphA4mRNA表達(dá)(分別為:0.69±0.11、0.67±0.09)均低于對照組大鼠(0.99±0.01)(均為P0.05);恢復(fù)28天組大鼠下丘EphA4mRNA表達(dá)(0.88±0.04)與對照組大鼠比較差異無統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論長期注射水楊酸鈉后大鼠下丘的EphA4表達(dá)呈可逆性下降,且與水楊酸鈉注射后大鼠聽功能損傷的變化一致,推測下丘神經(jīng)元軸突中EphA4參與了水楊酸鈉耳毒性的機(jī)制。
[Abstract]:Objective to investigate the role of EphA4 expression in sodium salicylate ototoxicity by observing the changes of EphA4mRNA expression in ABR and inferior colliculus after long-term sodium salicylate treatment in rats. Methods Thirty healthy adult male SD rats were randomly divided into 5 groups, 6 rats in each group: normal control group (no treatment), intramuscular injection of sodium salicylate (175 mg / kg, twice a day, 8 hours interval for 7 days), intramuscular injection of sodium salicylate (175mg / kg), intramuscular injection of sodium salicylate (175mg / kg). The rats were given intramuscular injection of sodium salicylate for 14 days (sodium salicylate for 14 days), recovery group for 14 days after sodium salicylate injection for 14 days, and group for 28 days after sodium salicylate was injected intramuscularly. The rats in each group were killed after ABR detection at the corresponding time points and the expression of EphA4mRNA in the inferior colliculus was detected by real-time fluorescence quantitative PCR (real-time PCR). Results 1the ABR response threshold of the rats in the group of intramuscular injection for 14 days was 38.33 鹵3.73 鹵44.16 鹵1.86dB SPL, which was significantly higher than that in the control group (30.83 鹵1.86dB SPL) (P0.05). There was no significant difference in ABR response threshold (32.50 鹵2.50dB SPL) compared with control group (30.83 鹵1.86dB SPL) after 28 days of recovery (P0.05). 2the expression of EphA4mRNA in the inferior colliculus of the rats in the 7-day group and the 14-day group (0.69 鹵0.110.67 鹵0.09, respectively) was lower than that in the control group (0.99 鹵0.01) (P0.05); The expression of EphA4mRNA in inferior colliculus of rats in 28 days group (0.88 鹵0.04) was not significantly different from that in control group (P0.05). Conclusion the expression of EphA4 in the inferior colliculus of rats after long-term injection of sodium salicylate was inversely decreased, which was consistent with the changes of auditory function of rats after sodium salicylate injection. It is speculated that EphA4 in axons of inferior colliculus neurons may play a role in the mechanism of sodium salicylate ototoxicity.
