8-OHdG和VEGF在原發(fā)性翼狀胬肉組織中的表達(dá)及意義

【摘要】:目的通過檢測(cè)8-羥基脫氧鳥苷(8-OHd G)及血管內(nèi)皮生長(zhǎng)因子(VEGF)在原發(fā)性翼狀胬肉和正常結(jié)膜組織中的表達(dá),探討DNA氧化損傷及新生血管形成在原發(fā)性翼狀胬肉發(fā)病機(jī)制中的作用。方法收集手術(shù)切除的原發(fā)性翼狀胬肉組織標(biāo)本35例(35眼),并收集術(shù)眼顳上方正常球結(jié)膜標(biāo)本5例(5眼)作對(duì)照。分別采用免疫組織化學(xué)法和實(shí)時(shí)熒光定量PCR技術(shù)檢測(cè)翼狀胬肉和結(jié)膜組織標(biāo)本中8-OHd G及VEGF的表達(dá),分析比較兩種因子在不同組織間的表達(dá)差異及表達(dá)規(guī)律。結(jié)果1.8-OHd G在翼狀胬肉和結(jié)膜組織標(biāo)本中的表達(dá)差異及規(guī)律在35例翼狀胬肉組織中有24例呈陽性表達(dá),陽性表達(dá)率為68.6%,而正常結(jié)膜組織中無8-OHd G的表達(dá),其陽性表達(dá)率的差異具有顯著統(tǒng)計(jì)學(xué)意義(P=0.007)。8-OHd G的陽性表達(dá)位于胬肉組織上皮細(xì)胞的細(xì)胞核、呈棕黃色著染,細(xì)胞質(zhì)無染色。上皮下的纖維血管組織及正常結(jié)膜組織無表達(dá)。2.VEGF在翼狀胬肉和結(jié)膜組織標(biāo)本中的表達(dá)差異及規(guī)律(1)VEGF m RNA在翼狀胬肉各部位及正常球結(jié)膜(1.18323±0.702762)中均有表達(dá),且自翼狀胬肉頭部(0.99526±0.525440)、頸部(1.38062±0.715145)至體部(2.41824±0.906681)表達(dá)量呈逐漸增高趨勢(shì)。(2)VEGF m RNA在翼狀胬肉體部的表達(dá)量明顯高于頭部和頸部,表達(dá)的差異均具有顯著的統(tǒng)計(jì)學(xué)意義(P0.001)。(3)翼狀胬肉體部VEGF的表達(dá)較正常結(jié)膜明顯增高,差異具有顯著統(tǒng)計(jì)學(xué)意義(P=0.0020.01);但頭部、頸部與正常結(jié)膜三者之間的表達(dá)差異無明顯統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論1.8-OHd G及VEGF在原發(fā)性翼狀胬肉組織中的表達(dá)均比正常結(jié)膜中表達(dá)明顯增高,提示DNA氧化應(yīng)激損傷及新生血管形成在原發(fā)性翼狀胬肉發(fā)病機(jī)制中發(fā)揮重要作用。2.VEGF在原發(fā)性翼狀胬肉各部位的表達(dá)自頭部至體部呈逐漸增高趨勢(shì),提示翼狀胬肉的新生血管過度增生位于胬肉體部。
[Abstract]:Objective to detect the expression of 8-hydroxydeoxyguanosine (8-OHd G) and vascular endothelial growth factor (VEGF) in primary pterygium and normal conjunctiva. To investigate the role of DNA oxidative injury and angiogenesis in the pathogenesis of primary pterygium. Methods 35 cases (35 eyes) of primary pterygium were collected and 5 cases (5 eyes) of normal bulbar conjunctiva were collected as control. The expression of 8-OHd G and VEGF in pterygium and conjunctiva was detected by immunohistochemistry and real-time fluorescence quantitative PCR. Results the expression of 1.8-OHd G in pterygium and conjunctiva tissues was different and regular. 24 cases of pterygium showed positive expression in 35 cases of pterygium, the positive rate was 68.6%, but there was no expression of 8-OHd G in normal conjunctiva. The positive expression rate of 8-OHd G was located in the nucleus of pterygium epithelium cells and was stained with brown color and no staining in cytoplasm. The expression of 2.VEGF in pterygium and conjunctiva tissues was different (1) VEGF m RNA was expressed in all parts of pterygium and normal bulbar conjunctiva (1.18323 鹵0. 0). 702762). The expression of pterygium was increased from head (0.99526 鹵0.525440), neck (1.38062 鹵0.715145) to body (2.41824 鹵0.906681). (2) the expression of pterygium in pterygium was significantly higher than that in head and neck. The expression of VEGF in pterygium was significantly higher than that in normal conjunctiva (P0. 001). (3), and the difference was statistically significant (P0. 0020.01). However, there was no significant difference between head, neck and normal conjunctiva (P0.05). Conclusion the expression of 1.8-OHd G and VEGF in primary pterygium is significantly higher than that in normal conjunctiva. The results suggest that oxidative stress injury and angiogenesis of DNA play an important role in the pathogenesis of primary pterygium. The expression of 2.VEGF in all parts of primary pterygium is increasing gradually from head to body. The results suggest that the hyperplastic neovascularization of pterygium is located in pterygium.
