氟西汀聯(lián)合對(duì)側(cè)眼遮蓋對(duì)成年大鼠弱視的治療作用

【摘要】:目的研究氟西汀(fluoxetine,FLx)口服配合傳統(tǒng)遮蓋治療對(duì)成年大鼠弱視的治療效果。設(shè)計(jì)實(shí)驗(yàn)研究。研究對(duì)象成年弱視大鼠(P70)。方法制作大鼠成年弱視模型,根據(jù)干預(yù)方式不同將弱視大鼠隨機(jī)分為對(duì)照組(正常飲水喂養(yǎng))、Flx2組(20%FLx水喂養(yǎng)2周)、Flx4組(20%FLx水喂養(yǎng)4周)、BFD組(雙眼形覺(jué)剝奪2周),各組大鼠行傳統(tǒng)非弱視眼遮蓋(翻轉(zhuǎn)縫合眼)治療后行圖形視覺(jué)誘發(fā)電位(PVEP)檢查。主要指標(biāo)PVEP振幅,對(duì)側(cè)眼與同側(cè)眼振幅比值(the ratio of contralateral versus ipsilateral,C/I比值)及組間比較的HSD-Q值。結(jié)果 Flx4組及BFD組配合非弱視眼遮蓋治療1周后弱視眼PVEP振幅由(19.86±3.07)μv、(17.51±4.12)μv升高至(32.31±4.29)μv、(29.31±2.29)μv(t=-4.938、-4.301,P=0.001、0.001);C/I值由1.11±0.29、1.04±0.28提高至2.34±0.45、2.45±0.29(t=-2.743、-3.629,P=0.016、0.007);遮蓋治療后C/I值組間差異顯著(F=18.86,P=0.018;對(duì)照組與Flx2組:HSD-Q=0.13,P=0.175;Flx2組與Flx4組:HSD-Q=9.76,P=0.013;Flx4組與BFD組:HSD-Q=0.09,P=0.350);BFD組還出現(xiàn)健眼PVEP振幅下降(t=2.672,P=0.047)。FLx2組及對(duì)照組弱視眼PVEP振幅無(wú)明顯提高,C/I比值未出現(xiàn)明顯變化。結(jié)論20%氟西汀水溶液口服4周與雙眼形覺(jué)剝奪2周均可穩(wěn)定激活成年弱視大鼠視皮層可塑性;氟西汀口服配合非弱視眼遮蓋治療可成為成年大鼠弱視的治療手段。
