【摘要】：目的:通過(guò)對(duì)慢性扁桃體炎患者灼燒技術(shù)治療前后TLR-NF-κB信號(hào)通路中的重要參與物質(zhì)及部分相關(guān)細(xì)胞因子的濃度與健康志愿者相同指標(biāo)的濃度進(jìn)行比較研究,探討該治療方法對(duì)慢性扁桃體炎患者免疫功能所產(chǎn)生的影響和作用機(jī)制。方法:選取于我院2016年4月至2016年10月間門診就診并診斷為慢性扁桃體炎的患者20例為試驗(yàn)組,將其在灼燒技術(shù)治療前以及一療程灼燒技術(shù)治療后采靜脈血,將所得標(biāo)本離心分離后獲得血清,然后采用酶聯(lián)免疫吸附試驗(yàn)檢測(cè)TLR-NF-κB信號(hào)通路中TLR2、TLR4、NF-κB在外周血的濃度,同時(shí)檢測(cè)細(xì)胞因子IL-4、IL-5、IL-10、IFN-γ及TGF-β在外周血的濃度,將患者治療前后以上指標(biāo)的濃度進(jìn)行對(duì)比。同時(shí)招募健康志愿者6例作為對(duì)照組,靜脈采血后分離得到血清,并采用相同方法檢測(cè)以上指標(biāo)的濃度,將試驗(yàn)組灼燒治療前患者血清中以上指標(biāo)的表達(dá)情況與對(duì)照組進(jìn)行比較。所得數(shù)據(jù)采用SPSS21.0進(jìn)行統(tǒng)計(jì)分析。結(jié)果:將試驗(yàn)組灼燒治療前血清中所檢測(cè)指標(biāo)的濃度與對(duì)照組的進(jìn)行比較,前者血清中TLR2、TLR4、NF-κB、IL-4、IL-5、IL-10、IFN-γ及TGF-β的濃度明顯高于對(duì)照組,差異有統(tǒng)計(jì)學(xué)意義(P0.05);將試驗(yàn)組患者灼燒治療前與治療后進(jìn)行比較,慢性扁桃體炎患者灼燒治療后其血清中TLR2、TLR4、IL-4、IL-5、IL-10、NF-κB、IFN-γ及TGF-β的濃度比治療前低,差異有統(tǒng)計(jì)學(xué)意義(P0.05);將試驗(yàn)組患者灼燒治療前血清中TLR2、TLR4、IL-4、IL-5、IL-10、IFN-γ、TGF-β與NF-κB進(jìn)行關(guān)聯(lián)性分析,統(tǒng)計(jì)分析表明以上檢測(cè)指標(biāo)與NF-κB不存在關(guān)聯(lián)性(P0.05)。而將慢性扁桃體炎患者灼燒治療后血清中TLR2、TLR4、IL-4、IL-5、IL-10、IFN-γ、TGF-β與NF-κB進(jìn)行關(guān)聯(lián)性分析,統(tǒng)計(jì)分析表明灼燒治療后血清中TLR2與NF-κB的呈正相關(guān),而其他檢測(cè)指標(biāo)不存在關(guān)聯(lián)性(P0.05)。結(jié)論:血清中TLR2、TLR4、IL-4、IL-5、IL-10、NF-κB、IFN-γ、TGF-β均參與了慢性扁桃體炎患者疾病的發(fā)生與發(fā)展,灼燒技術(shù)對(duì)該病的治療作用與干預(yù)上述指標(biāo)的表達(dá)關(guān)系密切,該治療方式治療慢性扁桃體炎的機(jī)制與干預(yù)TLR-NF-κB信號(hào)通路及該路徑的部分細(xì)胞因子相關(guān),且與TLR2所參與的TLR-NF-κB信號(hào)通路關(guān)系密切。
[Abstract]:Objective: to compare the concentrations of important involved substances and some related cytokines in TLR-NF- 魏 B signaling pathway between patients with chronic tonsillitis and healthy volunteers before and after burn therapy. To investigate the effect and mechanism of this treatment on immune function in patients with chronic tonsillitis. Methods: twenty patients with chronic tonsillitis from April 2016 to October 2016 in our hospital were selected as experimental group. Venous blood was collected before burning and after one course of burning. The serum was obtained by centrifugation. The concentration of TLR2,TLR4,NF- 魏 B in peripheral blood and the concentration of cytokines IL-4,IL-5,IL-10,IFN- 緯 and TGF- 尾 in peripheral blood of TLR-NF- 魏 B signaling pathway were detected by enzyme-linked immunosorbent assay (Elisa). The concentrations of the above indexes were compared before and after treatment. At the same time, 6 healthy volunteers were recruited as the control group, the serum was isolated after venous blood sampling, and the concentration of the above indexes was detected by the same method. The expression of the above indexes in serum of patients in the experimental group before burn treatment was compared with that in the control group. The data were analyzed by SPSS21.0. Results: the serum concentrations of TLR2,TLR4,NF- 魏 B, IL-4, IL-5, IL-10, IFN- 緯 and TGF- 尾 in the experimental group were significantly higher than those in the control group before burn treatment. The difference was statistically significant (P0.05). The serum levels of TLR2,TLR4,IL-4,IL-5,IL-10,NF- 魏 B, IFN- 緯 and TGF- 尾 in patients with chronic tonsillitis were lower than those before and after treatment. The correlation between TLR2,TLR4,IL-4,IL-5,IL-10,IFN- 緯 TGF- 尾 and NF- 魏 B was analyzed before burn treatment in the experimental group (P0.05). The statistical analysis showed that the above indexes were not correlated with NF- 魏 B (P0.05). The correlation between TLR2,TLR4,IL-4,IL-5,IL-10,IFN- 緯 TGF- 尾 and NF- 魏 B in serum of patients with chronic tonsillitis after burn treatment was analyzed. The statistical analysis showed that there was a positive correlation between TLR2 and NF- 魏 B in serum after burn treatment, but there was no correlation between TLR2 and NF- 魏 B in serum of patients with chronic tonsillitis (P0.05). Conclusion: the serum TLR2,TLR4,IL-4,IL-5,IL-10,NF- 魏 B of IFN- 緯 TGF- 尾 is involved in the occurrence and development of chronic tonsillitis. The therapeutic effect of burning is closely related to the expression of the above indexes. The mechanism of the treatment of chronic tonsillitis is related to the interference of TLR-NF- 魏 B signaling pathway and some cytokines of this pathway, and is closely related to the TLR-NF- 魏 B signaling pathway involved by TLR2.
