【摘要】：目的炎癥在慢性鼻-鼻竇炎伴鼻息肉(chronic rhinosinusitis with nasal polyps,CRSwNP)的發(fā)生發(fā)展過程中起著重要作用,并且炎癥在CRSwNP的病理機(jī)制仍然不完全清楚。此研究主要觀察清道夫受體CD36在慢性鼻-鼻竇炎伴鼻息肉的表達(dá)。方法使用逆轉(zhuǎn)錄酶聚合酶鏈反應(yīng)(RT-PCR),Western blot免疫印跡方法檢測來自合計(jì)43例慢性鼻-鼻竇炎(chronic rhinosinusitis,CRS)或伴息肉患者的手術(shù)標(biāo)本CD36的mRNA和蛋白表達(dá)。進(jìn)一步通過免疫組織化學(xué)和免疫熒光技術(shù)觀察CD36表達(dá)與分布規(guī)律。結(jié)果我們的RT-PCR和Western blot免疫印跡分析的結(jié)果表明,與正常鼻黏膜組織相比,鼻息肉標(biāo)本中CD36 mRNA和蛋白表達(dá)顯著增強(qiáng)(P0.01)。免疫組織化學(xué)結(jié)果顯示,CD36弱表達(dá)于慢性鼻竇炎鼻黏膜組織,而鼻息肉組織中表達(dá)較強(qiáng)。CD36與血管內(nèi)皮標(biāo)記物CD31的雙染熒光結(jié)果顯示,CD36陽性的細(xì)胞與CD31的陽性共標(biāo)。結(jié)論研究數(shù)據(jù)表明CD36在慢性鼻-鼻竇炎伴鼻息肉發(fā)生與發(fā)展過程可能起著重要作用。
[Abstract]:Objective inflammation plays an important role in the development of chronic rhinosinusitis with nasal polyp (chronic rhinosinusitis with nasal polyps,CRSwNP), and the pathological mechanism of inflammation in CRSwNP is still unclear. The purpose of this study was to investigate the expression of scavenger receptor CD36 in chronic rhinosinusitis with nasal polyps. Methods the expression of mRNA and protein in CD36 from 43 patients with chronic rhinosinusitis (chronic rhinosinusitis,CRS) or polyps was detected by reverse transcriptase polymerase chain reaction (RT-PCR) Western blot Western blot. The expression and distribution of CD36 were observed by immunohistochemistry and immunofluorescence technique. Results the results of RT-PCR and Western blot Western blot analysis showed that the expression of CD36 mRNA and protein in nasal polyps was significantly higher than that in normal nasal mucosa (P0.01). The results of immunohistochemistry showed that the expression of CD36 was weak in the nasal mucosa of chronic sinusitis, while the expression of CD36 in nasal polyps was stronger than that in the vascular endothelial marker CD31. The results showed that the cells with positive expression of CD36 were colabeled with CD31. Conclusion CD36 may play an important role in the occurrence and development of chronic rhinosinusitis with nasal polyps.
