microRNA-126在多能干細(xì)胞定向分化為視網(wǎng)膜神經(jīng)干細(xì)胞過(guò)程中表達(dá)量的變化及意義

【摘要】:目的研究microRNA-126(miR-126)在濕性老年黃斑變性(wet AMD)疾病患者來(lái)源的多能干細(xì)胞(i PSC)定向分化為視網(wǎng)膜神經(jīng)干細(xì)胞(RNSC)中表達(dá)量的變化及意義。方法選取新發(fā)現(xiàn)的wet AMD患者10例作為觀察組,無(wú)眼底病變及全身病的10例作為對(duì)照組。抽取受試者的外周靜脈血,提取單個(gè)核細(xì)胞(PBMC),進(jìn)一步提取CD34+細(xì)胞,行腺病毒轉(zhuǎn)染,誘導(dǎo)產(chǎn)生i PSC且進(jìn)行鑒定;應(yīng)用實(shí)時(shí)定量PCR(qRT-PCR)技術(shù)檢測(cè)miR-126在10個(gè)不同誘導(dǎo)發(fā)育階段的RNSC中的相對(duì)表達(dá)量。結(jié)果 wet AMD疾病患者的外周血成功誘導(dǎo)分化出RNSC;觀察組miR-126的不同誘導(dǎo)時(shí)間相對(duì)表達(dá)量的差異除了時(shí)刻8、9、10其余測(cè)試時(shí)間點(diǎn)均有統(tǒng)計(jì)學(xué)意義(P0.05);對(duì)照組miR-126的不同誘導(dǎo)時(shí)間相對(duì)表達(dá)量的差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05);觀察組與對(duì)照組細(xì)胞miR-126相對(duì)表達(dá)量在不同誘導(dǎo)時(shí)間的差異有統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論 wet AMD患者早期miR-126的表達(dá)量顯著升高,且有逐漸降低的趨勢(shì)。
[Abstract]:Objective to study the changes and significance of microRNA-126 (miR-126) expression in the differentiation of multipotent stem cells (i PSC) into retinal neural stem cells (RNSC) in elderly patients with (wet AMD) disease of macular degeneration. Methods Ten newly discovered wet AMD patients were selected as observation group, 10 patients without fundus disease and 10 patients with systemic disease as control group. The peripheral venous blood was extracted and the (PBMC), of mononuclear cells was extracted. The CD34 cells were further extracted by adenovirus transfection and I PSC was induced and identified. Real time quantitative PCR (qRT-PCR) was used to detect the relative expression of miR-126 in 10 RNSC at different stages of induction and development. Results RNSCs were successfully induced and differentiated from peripheral blood of patients with wet AMD disease, and the difference of relative expression of miR-126 at different induction time in observation group was statistically significant (P0.05), and that in control group was different in different miR-126 induction time (P0.05). There was no significant difference in the relative expression of miR-126 between the two groups (P0.05); there was significant difference in the relative expression of miR-126 between the observation group and the control group at different induction time (P0.05). Conclusion the expression of miR-126 in the early stage of wet AMD patients increased significantly and tended to decrease gradually.
