甘蔗葉多糖對鼻咽癌細(xì)胞株CNE2的體外抑制作用研究

【摘要】:目的:研究甘蔗葉多糖對鼻咽癌CNE_2細(xì)胞株的體外抑制作用,并初步探討其作用機(jī)制。 方法:采用熱水浸提及DEAE-纖維素層析柱法提純甘蔗葉多糖,以MTT法觀察其體外抗腫瘤活性,透射電子顯微鏡觀察凋亡細(xì)胞的超微結(jié)構(gòu),采用流式細(xì)胞術(shù)檢測腫瘤細(xì)胞凋亡率,實(shí)時(shí)熒光定量PCR法檢測凋亡相關(guān)基因Bax、Bcl-2的變化。 結(jié)果:濃度為10ug/ml、20ug/ml、40ug/ml、80ug/ml、160ug/ml的甘蔗葉多糖作用后,CNE_2細(xì)胞抑制率及凋亡率均明顯增加,且呈濃度依賴性。電鏡下可見160ug/ml濃度的甘蔗葉多糖可使CNE_2細(xì)胞出現(xiàn)凋亡小體。熒光定量PCR技術(shù)測定Bax在CNE_2細(xì)胞中表達(dá)上調(diào),并隨著甘蔗葉多糖作用濃度的增加而升高;Bcl-2則在CNE_2細(xì)胞中表達(dá)下調(diào),,且隨著甘蔗葉多糖作用濃度的增加而降低。 結(jié)論:甘蔗葉多糖能明顯抑制人鼻咽癌CNE_2細(xì)胞增殖,誘導(dǎo)細(xì)胞凋亡,可能通過提高Bax表達(dá)及降低Bcl-2的表達(dá),從而發(fā)揮抗腫瘤作用。
[Abstract]:Aim: to study the inhibitory effect of sugarcane leaf polysaccharide on CNE2 cell line of nasopharyngeal carcinoma (NPC) in vitro and to explore its mechanism. Methods: the polysaccharides from sugarcane leaves were purified by hot water extraction and DEAE- cellulose chromatography. The in vitro antitumor activity was observed by MTT assay, the ultrastructure of apoptotic cells was observed by transmission electron microscope, and the apoptosis rate of tumor cells was detected by flow cytometry. The changes of apoptosis-related gene Baxon Bcl-2 were detected by real-time fluorescence quantitative PCR. Results: the inhibition rate and apoptosis rate of CNE2 cells were significantly increased after the treatment of sugar cane leaf polysaccharides at the concentration of 10ugrml / ml 20ugr / ml 40ugrml / ml ~ 80ugr / ml sugarcane leaf polysaccharide in a concentration-dependent manner. The inhibitory rate and apoptosis rate of CNE2 cells were increased in a concentration-dependent manner. Under electron microscope, 160ug/ml concentration of polysaccharides from sugarcane leaves could make CNE2 cells appear apoptotic bodies. The expression of Bax was up-regulated in CNE2 cells by fluorescence quantitative PCR, and the expression of Bcl-2 was down-regulated in CNE2 cells with the increase of polysaccharide concentration in sugarcane leaves, and decreased with the increase of polysaccharide concentration in sugarcane leaves. Conclusion: polysaccharide from sugarcane leaves can significantly inhibit the proliferation of CNE2 cells and induce apoptosis, which may play an anti-tumor role by increasing the expression of Bax and decreasing the expression of Bcl-2.
