白藜蘆醇誘導(dǎo)Hep-2人喉表皮樣癌細(xì)胞自噬作用

【摘要】:目的:觀察白藜蘆醇對Hep-2細(xì)胞形態(tài)學(xué)的影響及誘導(dǎo)其自噬的可能機(jī)制。方法:40μM濃度白藜蘆醇復(fù)合培養(yǎng)液作用于Hep-2細(xì)胞后,MDC染色和透射電鏡分別檢測自噬小體和自噬泡;Hoechest33258染色檢測凋亡細(xì)胞;western blot分別分析自噬基因蛋白水平。結(jié)果:白藜蘆醇增加LC3Ⅱ與LC3Ⅰ的比率,促進(jìn)beclin1蛋白和mRNA的表達(dá),細(xì)胞內(nèi)的自噬泡和自噬小體數(shù)量也顯著增加。用3MA和CQ抑制自噬則白藜蘆醇對Hep-2細(xì)胞的促凋亡作用顯著減弱。結(jié)論:白藜蘆醇能夠促進(jìn)Hep-2細(xì)胞自噬;白藜蘆醇誘導(dǎo)Hep-2細(xì)胞自噬的機(jī)制可能與促進(jìn)LC3Ⅱ與LC3Ⅰ的表達(dá)、增強(qiáng)自噬基因beclin1蛋白的表達(dá)有關(guān)。
[Abstract]:Aim: to observe the effect of resveratrol on the morphology of Hep-2 cells and the possible mechanism of inducing autophagy.
