川芎嗪對(duì)慢性高眼壓大鼠視網(wǎng)膜神經(jīng)節(jié)細(xì)胞凋亡的影響

【摘要】:目的研究川芎嗪對(duì)慢性高眼壓大鼠視網(wǎng)膜神經(jīng)節(jié)細(xì)胞(retina ganglion cells,RGC)凋亡的保護(hù)作用。方法健康SD大鼠30只,隨機(jī)分為正常組、高眼壓組和川芎嗪治療組,每組10只。高眼壓組和治療組分別采用上鞏膜靜脈燒灼法制作高眼壓模型,正常對(duì)照組不做處理。造模成功后7d治療組給予川芎嗪腹腔注射,正常組與高眼壓組給予等量的生理鹽水腹腔注射。造模3周后取三組大鼠眼球,行視網(wǎng)膜HE染色、TUNEL原位細(xì)胞凋亡檢測(cè)和Bcl-2免疫組織化學(xué)檢測(cè)。結(jié)果造模后高眼壓組、川芎嗪治療組各時(shí)間點(diǎn)眼壓與正常組比較,差異均有統(tǒng)計(jì)學(xué)意義(均為P0.05),而兩組間各時(shí)間點(diǎn)比較,差異均無(wú)統(tǒng)計(jì)學(xué)意義(均為P0.05)。正常組、高眼壓組、川芎嗪治療組大鼠視網(wǎng)膜RGC層TUNEL陽(yáng)性細(xì)胞數(shù)分別為0、9.00±0.89、3.50±1.89,三組間差異有統(tǒng)計(jì)學(xué)意義(F=86.160,P=0.000);三組間兩兩比較,差異均有統(tǒng)計(jì)學(xué)意義(均為P=0.000)。三組大鼠RGC層Bcl-2陽(yáng)性表達(dá)光密度值分別為0.373±0.090、0.375±0.146、0.390±0.268,三組差異有統(tǒng)計(jì)學(xué)意義(F=84.658,P=0.000);三間組兩兩比較,差異均有顯著統(tǒng)計(jì)學(xué)意義(均為P=0.000)。結(jié)論川芎嗪通過(guò)上調(diào)Bcl-2蛋白的表達(dá)可明顯抑制慢性高眼壓大鼠RGC凋亡,從而發(fā)揮RGC保護(hù)作用。
[Abstract]:Objective to study the protective effect of ligustrazine on apoptosis of retinal ganglion cells (RGCs) in rats with chronic intraocular hypertension. Methods 30 healthy Sprague-Dawley rats were randomly divided into normal group, intraocular hypertension group and ligustrazine treatment group with 10 rats in each group. The model of high intraocular pressure was made by scleral vein cauterization in the high intraocular pressure group and the treatment group, but not in the normal control group. The rats in the treatment group were intraperitoneally injected with ligustrazine, and the normal group and the high intraocular pressure group were intraperitoneally injected with the same amount of normal saline on the 7th day after the successful establishment of the model. After 3 weeks, the eyeballs of the three groups were taken out, and the in situ apoptosis and Bcl-2 immunohistochemistry were detected by HE staining and Tunel in the retina. Results there were significant differences in intraocular pressure (IOP) between the two groups at different time points (P 0.05), but there was no significant difference between the two groups at each time point (P 0.05). The number of Tunel positive cells in RGC layer of rats in normal group, high intraocular pressure group and ligustrazine treatment group was 0 9. 00 鹵0. 89 鹵0. 50 鹵1. 89, respectively. The difference among the three groups was statistically significant (P 0. 000). The positive optical density of Bcl-2 expression in RGCs of the three groups was 0.373 鹵0.090 鹵0.375 鹵0.146 鹵0.390 鹵0.268 respectively, and the difference among the three groups was statistically significant (P < 0.01). Conclusion Ligustrazine can obviously inhibit the apoptosis of RGC in rats with chronic intraocular hypertension by up-regulating the expression of Bcl-2 protein, thus exerting the protective effect of RGC.
