【摘要】：目的:目的是研究三七姜揮發(fā)油對人鼻咽癌CNE-2細(xì)胞侵襲和遷移的抑制作用,并探討其可能機制。方法:體外培養(yǎng)人鼻咽癌CNE-2細(xì)胞,CCK8實驗檢測三七姜揮發(fā)油對CNE-2細(xì)胞增值能力的抑制作用,計算生長抑制率及半數(shù)抑制濃度(IC50);選取三七姜揮發(fā)油(0,20,40,60μg·ml-1)作用于CNE-2細(xì)胞0,24,48h,分別于三個不同時間段,采用劃痕實驗檢測三七姜揮發(fā)油對CNE-2細(xì)胞遷移能力的抑制作用;Transwell實驗檢測三七姜揮發(fā)油對CNE-2細(xì)胞侵襲能力的抑制作用。取作用48h后的CNE-2細(xì)胞,采用蛋白印跡法檢測MMP-2、MMP-9和VEGF等侵襲和遷移相關(guān)基因蛋白水平的表達;RT-PCR技術(shù)檢測MMP-2、MMP-9和VEGF等相關(guān)侵襲和遷移基因的表達情況。結(jié)果:三七姜揮發(fā)油作用于CNE-2細(xì)胞24h,48h,72h后,細(xì)胞半數(shù)抑制濃度分別為70.296μg·ml-1,63.749μg·ml-1,59.532μg·ml-1,呈時間-劑量依賴性(P0.05);不同濃度的三七姜揮發(fā)油(0,20,40,60μg·ml-1)作用于細(xì)胞48h后,劃痕實驗發(fā)現(xiàn)與空白對照組相比,細(xì)胞的遷移能力隨著藥物濃度的增加而降低;Transwell實驗顯示,隨著藥物濃度逐漸的增加,細(xì)胞的侵襲能力逐漸下降;RT-PCR顯示,與陰性對照組相比,MMP-2、MMP-9和VEGF等基因表達下調(diào),且變化趨勢呈一定的濃度依賴性。蛋白免疫印跡法顯示,與陰性對照組相比,MMP-2、MMP-9和VEGF等蛋白表達下調(diào),且變化趨勢呈一定的濃度依賴性。結(jié)論:三七姜揮發(fā)油能抑制人鼻咽癌CNE-2細(xì)胞侵襲和遷移的能力而發(fā)揮抗腫瘤的作用,并且成濃度依賴性,其機制可能與通過下調(diào)MMP-2、MMP-9和VEGF等基因的表達,進而抑制鼻咽癌腫瘤細(xì)胞的轉(zhuǎn)移有關(guān)。
[Abstract]:Aim: to study the inhibitory effect of volatile oil of Panax notoginseng on invasion and migration of human nasopharyngeal carcinoma (NPC) CNE-2 cells and its possible mechanism. Methods: the inhibitory effect of essential oil of Panax notoginseng on the proliferation of human nasopharyngeal carcinoma (NPC) CNE-2 cells was detected by CCK8 assay. The growth inhibition rate and the half inhibitory concentration (IC50) were calculated, and the essential oil of Radix Notoginseng was selected. The inhibitory effect of essential oil of Panax notoginseng on the migration of CNE-2 cells was detected by scratch test. The inhibitory effect of volatile oil of Panax notoginseng on the invasion of CNE-2 cells was detected by Transwell test. The expression of MMP-2, MMP-9, VEGF and other invasion and migration-related genes were detected by Western blotting in CNE-2 cells after 48 h. RT-PCR was used to detect the expression of invasion and migration genes such as MMP-2, MMP-9, VEGF and so on. Results: after treated with the volatile oil of Panax notoginseng for 24 h and 48 h and 72 h after treatment, the cell half inhibitory concentrations were 70.296 渭 g ml-1nl and 63.749 渭 g ml-1n 59.532 渭 g ml-1, respectively, in a time-dose dependent manner (P0.05A), and different concentrations of volatile oil 020404060 渭 g ml-1) were applied to the cells for 48 h. The migration ability of cells decreased with the increase of drug concentration. With the increase of drug concentration, the invasiveness of cells decreased gradually. Compared with negative control group, the expression of MMP-2, MMP-9, VEGF and other genes were down-regulated by reverse transcription-polymerase chain reaction (RT-PCR). The trend of change was in a concentration dependent manner. Western blot showed that the expression of MMP-2, MMP-9 and VEGF were down-regulated in a concentration dependent manner. Conclusion: the essential oil of Panax notoginseng can inhibit the invasion and migration of human nasopharyngeal carcinoma CNE-2 cells and play an antitumor role in a concentration-dependent manner. The mechanism may be related to the down-regulation of the expression of MMP-2, MMP-9 and VEGF genes. And then inhibit the metastasis of nasopharyngeal carcinoma tumor cells.
