【摘要】：葡萄膜炎是一種反復(fù)發(fā)作的炎癥性疾病,可導(dǎo)致免疫系統(tǒng)功能障礙和多器官損傷.然而,葡萄膜炎是否導(dǎo)致肝功能損害尚不十分清楚.本文通過運(yùn)用流式分析技術(shù)和激光共聚焦成像技術(shù),研究了實(shí)驗(yàn)性自身免疫葡萄膜炎模型的肝臟病理和功能變化.結(jié)果顯示肝損傷可出現(xiàn)在葡萄膜炎的炎癥后期并與眼損傷程度相關(guān).并且CD3~+CD4~+T細(xì)胞、CD3~-NK1.1~+DX5~-NK細(xì)胞、和CD11b~~+F4/80~-ly6c~+細(xì)胞在感染的眼睛和肝臟中增加.將CD3~+CD4~+T細(xì)胞回輸給炎癥的小鼠后,眼睛和肝臟的病理損傷加重.此外,在炎癥的小鼠中可見血管擴(kuò)張,大量淋巴細(xì)胞浸潤到炎癥的眼和肝臟的血管周圍.總之,我們的研究結(jié)果提示,肝損傷可以發(fā)生在小鼠葡萄膜炎模型中,這種損傷可能與通過外周循環(huán)浸潤到肝臟的CD3~+CD4~+T細(xì)胞有關(guān).
[Abstract]:Uveitis is a recurrent inflammatory disease that can lead to dysfunction of the immune system and multiple organ damage. However, it is not clear whether uveitis leads to liver damage. The pathological and functional changes of liver in experimental autoimmune uveitis model were studied by flow analysis and confocal laser imaging. The results showed that liver injury could occur in the late stage of uveitis and correlated with the degree of ocular injury. And CD3 ~ CD4 ~ T cells, CD3 + -NK1.1 ~ DX5~-NK cells, and CD11bF4 / F4 / 80 ~ ly6c~ cells were increased in infected eyes and liver. The pathological damage of eyes and liver was aggravated after CD3 ~ CD4 ~ T cells were infused into inflammatory mice. In addition, vasodilation is seen in inflammatory mice, with large numbers of lymphocytes infiltrating the inflammatory eyes and around the blood vessels of the liver. In conclusion, our results suggest that liver injury may occur in mice with uveitis, which may be related to CD3 ~ CD4 ~ T cells infiltrating into the liver through peripheral circulation.
